Chromatography

Aim: Extract and identify the pigments in the bud and stalk of a dandelion

Research question: What pigments are contained in the bud and stalk of a dandelion?

Background information:

- Chromatography is a laboratory technique for the separation of a mixture. A solvent is

a substance that has can dissolve a mixture. Solubility is the property of a substance to

be dissolved in a solvent. A chromatography paper is a kind of paper used for

chromatography. Pigment is any group of compounds that is intensely coloured.

Hypothesis:

o I think that the only pigment I will be only to identify will be chlorophyll a or b since

the bud isn’t fully grown, and chlorophylls are the most common pigments found in

green plants.

Variables:

o Independent variables

o Plant used – this variable can completely change the result as different

types if plants have different types of pigments

o Dependent variables

o Retention values - this variable is affected by the solvent used on the plant,

since not every plant can be dissolved by every solvent and if the plant is not

properly dissolved, then the pigments won’t come out properly leading to bad

retention values

o Controlled variables

o Amount of solvent used – This variable can affect the retention factor

o Solvent – this variable directly affects what pigment in it will be extracted as

not every solvent can dissolve every plant

Materials:

o Chromatography paper

o Mortar and pestle

o Beaker

o 50ml of methanol

o Bud and stalk of dandelion

Procedure:

1. Crush the plant up with a small amount of solvent for about 5 minutes

2. Take everything that is still solid out

3. Draw a line 3cm from the bottom of the chromatography paper

4. Pour 5 drops of the mixture on the line

5. Fix the paper in the beaker

6. Pour the rest of the solvent in the beaker making sure it does not go over the line

7. Wait for 3 hours

8. Take the chromatography paper out and let it dry

9. Measure and note the travel distance of the solvent and pigment

Rf value: L2 / L1 = 15.2cm / 12.5 cm = 0.8

This value and colour correspond to the pigment pheophytin1.

Uncertainties:

- Ruler – 0.1 cm

Evaluation and conclusion:

- I am pretty satisfied with the results of my experiment as the pigment matches

the RF value and colour. There were a few errors namely: the colours my eyes

perceive are not 100% correct (random error), the chromatography paper

folded and wrinkled in the beaker (random error),
