ACTIVITY

ENUMERATION OF MICROORGANISMS FROM THE ENVIRONMENT

Microorganism enumeration from the environment is a scientific activity that aims to measure and identify the

bacterial load or concentration present in a particular sample. This method enables researchers to acquire

significant insights on the microbial composition of different settings as well as analyze the diversity and features

of bacterial strains.

Scientists obtain significant insights into the microbial world through the enumeration of microorganisms from the

environment, which contributes to sectors such as environmental monitoring, public health, and microbiology

research. This activity promotes a greater understanding of the diversity and dispersion of bacteria in various

habitats, ultimately leading to breakthroughs in several scientific areas.

I. Objectives:

● To quantify the bacterial load or concentration present in the sample.


● To identify the bacteria, present in the sample.
● To identify and classify bacterial strains present in the sample.

II. Materials:

1 tbsp unflavored gelatin (White)


500 mL distilled water
Sterilized Petri Dish

¼ Beef broth cube
III. Procedures:

PART I. CULTURE MEDIA


1. Prepare a petri dish with agar medium (sterilized petri dishes)
2. In the beaker, combine 1 tablespoon gelatin and ¼ broth cube.
3. Stir occasionally until boil
4. Put a label in the petri dish (A, B, C, D, E, F)
o Petri Dish A: Exposed to the air for 5 minutes
o Petri Dish B: Use cotton swab to wipe an area of door knob and
streak onto entire surface
o Petri Dish C: Fingers were touched to the hair for 5 seconds and
streak to the gelatin
o Petri Dish D: Coughed in the agar
o Petri Dish E: Fingers were touched to the shoes for 5 seconds and
streak to the gelatin
o Petri Dish F: Use cotton swab to wipe an area of cellphone and
streak to the gelatin
5. Cover the lid and incubate the culture. Let colonies form.
6. Place the slide under the microscope and observe the bacteria.

PART II. DRAWING AND MICROSCOPY

1. Draw the colonies that formed in culture media.

2. Microscopy: Draw the microbial cell/s present in a colony.
