(Rona Aybay, 1998) 1 ABSTRACT In this study, the representation and presentation of gender roles in children's magazines published between 1939-1945 is reviewed through the emphasis and theme of being a nation. The most important way to analyse the concept of gender, which has diversified and shaped in a cultural and historical context, is to review the entire field from social dynamics to political power patterns. This period, which can be described as the maturation years of the Republic and the transition to the nation-state process, is significant in terms of bringing the consciousness of "being a citizen" to the forefront, in the shadow of the Second World War that shaped the World and where one-party political life prevailed. In this context, it is investigated how gender identities, which cannot be positioned separately from the nationstate building policies, are constructed and through which themes are transferred to the young generation in the children's magazines that set out with the aim of creating an "ideal nationideal generation" during the building process of the nation-state. From this point of view, the genres such as stories, tales, poems, proses and jokes etc., which were published in the magazines Çocuk Sesi-Voice of Children (1939), Yavru Türk-Turkish Juveniles (1939),-Steady Children (1940),-Friend (1941), Bilmece-Puzzle (1943),-Week (1943),-(1943), Türk-Turkish Kid (1944) and Çocuk-Jolly Children (1945), were analysed in the qualitative analysis method within the framework of themes determined. According to the findings of the study, it is seen that the themes of femininity and masculinity are included in the content of the magazine on the axis of national and human values in the content for children. While the years chosen as a period when the Republic was settled and Turkish modernization was adopted, are increasing the content aimed at fostering and perpetuating the love of homeland in children, they construct the ideal roles of femininity and masculinity on the strong nation theme.

Bu calismada hizla gelisen iletisim teknolojileri ile toplumda bircok yonden dezavantajli olarak gorulen ve “dijital gocmen” olarak tanimlanan yasli bireylerin iliskisi incelenmektedir. Bu baglamda 60-74 yas arasi yasli bireylerin yeni iletisim teknolojilerini kullanim becerileri, kullanim amaclari, araclari ve sikliklari uzerinden degerlendirilmektedir. Bu amacla TUIK’in 2016 yili Hanehalki Bilisim Teknolojileri Kullanim Arastirmasi veri seti birincil analize tabi tutulmus ve ANOVA testi yapilmistir. Calismanin bulgularina gore yasli bireylerin internet teknolojileri ve platformlarina onemli olcude hakim olduklari tespit edilmekle birlikte dijital yeterlilik duzeyleri uzerinde demografik unsurlarin da etkili oldugu sonucuna ulasilmistir.

In this study, internet regulations were discussed as digital surveillance practices in the context of concepts such as "risk" "supervision" and "control" based on Lyon's "Surveillance Society" theory. At hermostable alkaline protease was isolated from the Aspergillus niger Z1 strain in a liquid Czapek Dox medium, containing casein (1% w/v) as the sole nitrogen source. Enzyme extract was subjected to electrophoresis in SDS-polyacrylamide gel. Two protein bands were seen on polyacrylamide gel. Active enzyme was visualized in a zymo- gram and protease activity exhibited a molecular mass of 68 kDa on SDS-polyacrylamide gel. The optimum pH for activity was found to be 9.0. The temperature optima of the enzyme was found to be 40 °C at pH 9.0 and it remained stable up to 90 °C, with 48.4% of the orig- inal activity retained after heat treatment at 90 °C for 15 min. Proteolytic activity was inhib- ited by PMSF, but slightly inhibited by SDS.

This paper reports the isolation and identification of chitinase-producing Bacillus from chitin-containing wastes, production of a thermostable and alkaline chitinasese, and enzyme characterization. Bacillus thuringiensis subsp. kurstaki HBK-51 was isolated from soil and was identified. Chitinase was obtained from supernatant of B. thuringiensis HBK-51 strain and showed its optimum activity at 110°C and at pH 9.0. Following 3 hours of incubation period, the enzyme showed a high level of activity at 110°C (96% remaining activity) and between pH 9.0 and 12.0 (98% remaining activity). Considering these characteristics, the enzyme was described as hyperthermophile-thermostable and highly alkaline. Two bands of the enzyme weighing 50 and 125 kDa were obtained following 12% SDS-PAGE analyses.
