Catalase is a crucial enzyme of the antioxidant defense system responsible for the maintenance of cellular redox homeostasis. The aim of the present study was to evaluate the molecular regulation of catalase (Hdh-CAT) in stress physiology, innate immunity, testicular development, metamorphosis, and cryopreserved sperm of Pacific abalone. Hdh-CAT gene was cloned from the digestive gland (DG) of Pacific abalone. The 2894 bp sequence of Hdh-CAT had an open reading frame of 1506 bp encoding 501 deduced amino acids. Fluorescence in situ hybridization confirmed Hdh-CAT localization in the digestive tubules of the DG. Hdh-CAT was induced by different types of stress including thermal stress, H2O2 induction, and starvation. Immune challenges with Vibrio, lipopolysaccharides, and polyinosinic–polycytidylic acid sodium salt also upregulated Hdh-CAT mRNA expression and catalase activity. Tropomyosin (TPM) is a contractile protein responsible for muscle contraction through its actin-binding activity. The complete sequence of TPM in Haliotis discus hannai (Hdh-TPM) was 2160 bp, encoding 284 amino acids, and contained a TPM signature motif and a TPM domain. Gene ontology (GO) analysis based on the amino acid sequence predicted Hdh-TPM to have an actin-binding function in the cytoskeleton. The 3D analysis predicted the Hdh-TPM to have a coiled-coil α-helical structure. Phylogenetically, Hdh-TPM formed a cluster with other TPM/TPM1 proteins during analysis. The tissue-specific mRNA expression analysis found the higher expression of Hdh-TPM in the heart and muscles; however, during embryonic and larval development (ELD), the higher expression was found in the trochophore larvae and veliger larvae. To characterize the involvement of FMRFamide in the reproduction and larval development of Pacific abalone Haliotis discus hannai, an FMRFamide cDNA (Hdh-FMRF2) was cloned from the cerebral ganglion (CG). Fluorescence in situ hybridization and qRT-PCR were performed for functional characterization. The Pacific abalone Haliotis discus hannai is a highly commercialized seafood in Southeast Asia. The aim of the present study was to determine the antioxidant activity and oxidative stress-oriented apoptosis pathway in saccharides supplemented cryopreserved sperm of Pacific abalone. Cryopreserved sperm showed impaired antioxidant defenses due to the reduced mRNA abundance of antioxidant genes (CAT, Cu/Zn-SOD, Mn-SOD, GPx, GR, and BCL-2), apoptosis inhibitor (HSP70, and HSP90) gene, and enzymatic antioxidant activity compared to fresh sperm. Such impaired antioxidant defenses caused an increase in the mRNA expression of apoptosis genes (Bax, and Caspase-3), finally leading to apoptosis. The impaired antioxidant defense also increased O2•− production and lipid peroxidation (MDA) levels, which further accelerated apoptosis. As structural components of sperm, tektins are thought to play a fundamental role in sperm flagellar motility. In this study, Tektin-4 (Hdh-TEKT4) gene was successfully cloned and characterized from the testis tissue in Pacific abalone, Haliotis discus hannai. The full-length cDNA of Hdh-TEKT4 was 1,983 bp, with a coding region of 1,350 bp encoding 51.83 kDa putative protein of 449 deduced amino acids. Hdh-TEKT4 contains a tektin domain including a nonapeptide signature motif (RPGVDLCRD). As a multinational river, the Naf River flows into the Bay of Bengal in the Indian Ocean, between the Cox's Bazar district of Bangladesh and the Rakhine state of Burma. In a multidisciplinary approach, several experiments were carried out to understand the seasonal diversity of the phytoplankton community structure. A total of four layers of water was sampled from four depths in the Naf River during monsoon (September) and winter (December) of 2016. 41 species of phytoplankton were identified, and 3 different dominant groups (Cyanobacteria, Diatoms, and Dinoflagellates) were found. Diatoms and cyanobacteria alone were found to be most prevalent. Higher species diversity was observed in the monsoon season, with Synedra sp. (1.84×105 cells L-1, 18.76%) and winter with Microcystis sp. (1.41×105 cells L-1, 17.74%), respectively. The present investigation was conducted to assess the status of fish farming and livelihood of fish farmer in the Barisal Sadar Upazila, Southern Bangladesh for a period of ten months from March 2015 to January 2016. Well-structured questionnaire was applied for primary data collection from fish farmer. The survey revealed that average pond size was 0.12 ha with 85% of the farmers having ponds of single ownership. Majority farmer (94%) preferred poly-culture system where maximum pond was seasonal (84%). Most of the fish farmers were belonged to the age groups of 36 to 50 years (52%), represented by 80% Muslims.Majority farmer (68%) had tin-shed house where majority were taken health facilities from Upazila doctor (32%). Most of the farmer (55%) lived in joint families where majority family members (41%) were between 4 -5. The post stocking control mechanism of pond fish culture in Abyaynagar, Manirampur, Bagherpara and Chaugacha Upazila under Jessore district was observed from May to November 2016 by interviewing 200 fish farmers. It reflects, majority farmer (56%) cultured tilapia fish but only 3% cultured Shrimp/Prawn. The highest stocking density (63%) was found 100 to 200 fry/decimal having 0.5 to 2.0 cm (72%) fingerling size with directly grow out pond stocking (52%). About 58% framer used balance feed with broadcasting method in the morning feeding (77%) only. Most of the farmers did not exchange water (58%) and can’t culture properly (66%) due to lack of money. Only 7% farmers went to the government fisheries officials where 40% went to chemical shopkeepers for getting tips if any problems took place. It’s therefore, important to offer with institutional and organizational helps, credit and extension services for sustainable fish production in the Southwestern Bangladesh.

The present study was conducted to evaluate the livelihood status of the fishermen Engaged in Fishing in the Estuaries of Sundarbans during the period from January to December, 2014. The investigation was conducted on the Hariabhanga, Haringhata, Pasur and Shibsa estuarine area of the Sundarbans using a structured questionnaire. It was found that most of the fishermen were belonged to the age groups of 21 to 40 years (66%), represented by 76% Muslim. An experiment was conducted to assess the open water fish diversity and fishing activities in the Padma River in Rajshahi, Bangladesh from the period of February 2012 to January 2013. A total number of 76 fish species belonging to 9 orders, 22 families and 41 genera were recorded. The most dominant fish order was identified as Cypriniformes comprising 33% of total catch. About 27 endangered fish species were found in this area. Of the total fish species, 24% species were commonly found throughout the year. Twenty one species have been considered as threatened belonging to the Critically Endangered (33.33%), Vulnerable (10.60%) and Endangered (13.04%) of the total fish species. A total of 18 types of fishing gear (7 fishing nets, 6 fishing traps, 3 hooks and lines and 2 harpoons) were used for harvesting live fishes. The less availability of fishes is the major concern of declining biodiversity not only in the study area but also in the whole country.

Carbonic anhydrases (CAs) are universal zinc ion containing metalloenzymes that play a pivotal role in various physiological processes. In this study, a CA I (designated as Hdh CA I) was isolated and characterized from the mantle tissue of Pacific abalone, Haliotis discus hannai. The full-length cDNA sequence of Hdh CA I was 1,417-bp in length, encoding a protein of 337 amino acids with molecular weight of 37.58 kDa. Hdh CA I sequence possessed a putative signal peptide of 22 amino acids and a CA catalytic function domain. The predicted protein shared 94 and 78% sequence identities with Haliotis gigantea and Haliotis tuberculata CA I, respectively. Results of phylogenetic analysis indicated that Hdh CA I was evolutionarily close to CA I of H. gigantea and H. tuberculata with high bootstrap values. Groupers are mostly found in the reef-associated marine habitat, of which some are pelagic and others are demersal. Recently, a grouper species called Oblique-banded grouper, Epinephelus radiatus (Day, 1868) was newly reported while conducting research work on the availability of reef-associated fishes in St. Martin Island. This species had never been reported to occur not only from this coral reef area but also from the water area of Bangladesh. E. radiatus was easily identified following morphological traits, especially color pattern. The findings of the present study added new distributional range for this grouper species from Bangladeshi water.

Pacific abalone (Haliotis discus hannai) is a commercially important high valued molluscan species. Its wild population has decreased in recent years. It is widely cultured in Korea. Traditional breeding programs have been implemented for hatchery production of abalone seeds. To obtain more genetic information for its molecular breeding program, a high-density linkage map and quantitative trait locus (QTL) for three growth-related traits was constructed for Pacific abalone. F1 cross population with two parents were sampled to construct the linkage map using genotyping by sequencing (GBS). A total of 664,630,534 clean reads and 56,686 SNPs were generated and 3,345 segregating SNPs were used to construct a consensus linkage map. The map spanned 1,747.023 cM with 18 linkage groups and an average interval of 0.55 cM. QTL analysis revealed two significant QTL in LG10 on the consensus linkage map of each growth-related trait. The aim of the present study was to improve the sperm cryopreservation technique for this valuable species using an antifreeze protein III (AFPIII). The aim of the present study was to determine effects of cryopreservation on gene expression and post thaw sperm quality of Pacific abalone. Two ions, Na+ (459.1 ± 3.1 mM) and Cl– (515.9 ± 1.1 mM), were predominant in the seminal plasma (pH: 6.8 ± 0.1; osmolarity: 1,126 ± 3 mOsmL–1). In this study, the IGFBP7 gene was cloned and characterized from mantle tissues of H. discus hannai, and designated as Hdh IGFBP7. The full-length cDNA sequence transcribed from the Hdh IGFBP7 gene was 1519-bp long with an open reading frame of 720-bp corresponding to a putative polypeptide of 239 amino acids. The molecular mass of its mature protein was approximately 23.44 KDa with an estimated isoelectric point (pI) of 5.35, and it shared significant homology with IGFBP7 gene of H. madaka. The 921-bp full-length sequence of Hdh IGFBP-5 cDNA transcript had an open reading frame of 411 bp encoding a predicted polypeptide of 136 amino acids, sharing high sequence identities with IGFBP-5 of H. diversicolor. The deduced Hdh IGFBP-5 protein contained a putative transmembrane domain (13-35 aa) in the N-terminal region. It also possessed a signature domain of IGFBP protein family (IB domain, 45-120 aa).
