    Serine/threonine protein phosphatase 2A (PP2A) regulates several physiological processes such as the cell cycle, cell growth, apoptosis, and signal transduction. In this study, we examined the expression and role of PP2A Cα in adipocyte differentiation. PP2A Cα expression and PP2A activity decreased during adipocyte differentiation in C3H10T1/2 and 3T3-L1 cells and the expression of adipocyte marker genes such as PPARγ and adiponectin increased. To further clarify the role of PP2A Cα in adipocyte differentiation, we constructed PP2A knockdown cells by infecting C3H10T1/2 cells with a lentivirus expressing a shRNA specific for the PP2A Cα (shPP2A cells). Silencing of PP2A Cα in C3H10T1/2 cells dramatically stimulated adipocyte differentiation and lipid accumulation, which were accompanied by expression of adipocyte marker genes.     Lung cancer is one of the malignant tumors with greatest morbidity and mortality around the world. The keys to targeted therapy are discovery of lung cancer biomarkers to facilitate improvement of survival and quality of life for the patients with lung cancer. Translationally controlled tumor protein (TCTP) is one of the most overexpressed proteins in human lung cancer cells by comparison to the normal cells, suggesting that it might be a good biomarker for lung cancer. In the present study, the targeted efficacy of dihydroartemisinin (DHA) on TCTP expression in the A549 lung cancer cell model was explored. DHA could inhibit A549 lung cancer cell proliferation, and simultaneously up-regulate the expression of TCTP mRNA, but down-regulate its protein expression in A549 cells. In addition, it promoted TCTP protein secretion. Therefore, TCTP might be used as a potential biomarker and therapeutic target for non-small cell lung cancers.

    Cardiac troponin-I (cTnI) and -T (cTnT) are sensitive and specific markers of myocardial injury. However, the role of increased cTnI and cTnT in percutaneous coronary intervention (PCI)-related myocardial injury remains controversial. In this prospective, single-center and double-blind study, we aimed to determine the diagnostic and prognostic value of cTnI as well as cTnT (cTns) in PCI-related myocardial injury in a Chinese population. A total of 1,008 patients with stable angina pectoris and non-ST-segment elevation acute coronary syndrome were recruited. The levels of cTnI and cTnT were examined before and after PCI. All patients were followed up for 26±9 months to observe the incidence of major adverse cardiac events (MACEs).     To investigate the clinical outcome and application value of single stage circumferential surgery for irreducible old dislocation of the inferior cervical vertebra combined with tiny joint interlocking. From Mar. 2004 to Aug. 2010,18 patients with old dislocation of the inferior cervical vertebra, in which 16 showed double joint interlocking and the others 2 showed single by the three dimensional CT scan. MRI showed that all patients had intervertebral disc injury including intervertebral disc rupture (9 cases), intervertebral disc herniation (2 cases) and the fracture of inferior cervical vertebra accompanying with intervertebral disc rupture (7 cases).     To discuss the resection pseudoarthrosis for pelvic malignant tumors around acetabular. From May 1997 to June 2005, 25 patients with malignant tumors around acetabular were treated surgically with resection pseudoarthrosis. The series comprised 15 males and 10 females with an average age of 42 years old (range from 16 to 75 years old). There were 4 osteosarcomas, 12 chondrosarcomas, 1 Ewing's sarcoma, 1 neuroectodermal tumor, 1 myeloma, 1 malignant fibrohistiocytoma, 2 synovial sarcomas, and 3 metastases. Pseudoarthrosis was performed after resection of pelvic malignant tumors around acetabular. The affected side was protected postoperatively by skin traction with 2 - 3 kg weight for 6 to 8 weeks.     This study was aimed to investigate the effect of cord blood dendritic cells (DCs) on the in vitro proliferation capability, immunophenotype changes, level of secreted cytokines and activity against leukemia cells of the homologous cytokine-induced killer (CIK) cells. DCs and CIK cells were induced from cord blood mononuclear cells. They were co-cultured at the ratio of 1:5, and CIK cells from cord blood or DC-CIK cells from peripheral blood were cultured as controls.     To observe the long-term result of fibula grafting for reconstruction of the distal radius after giant cell tumor excision. From March 1994 to November 2004, 31 cases of fibula grafting for reconstruction of the distal radius for giant cell tumors performed were analysed. There were 12 males and 19 females. The patients were from 19 to 48 years old, and the mean age was 31 years. Twenty-four patients had Campanacci grade 3 lesions, and 7 patients had Campanacci grade 2 lesions. There were 6 cases of vascularized fibular grafting and 25 cases of non-vascularized fibular grafting. The data of 31 patients of periacetabular malignant tumors who had limb salvage surgery between January 1999 and December 2006 was retrospectively reviewed. There were 14 females and 17 males with a mean age of 53 years (range, 42-75 years). Twenty-four patients had chondrosarcomas, 4 patients had Ewing sarcomas, 1 patient had osteosarcoma, and the remaining 2 patients had metastatic disease. The retrospective analysis was performed on 83 patients undergoing the prosthetic reconstruction after the resection of the primary tumor around the knee between December 1995 and December 2005.     Bilateral pheochromocytomas are rare diseases. The purpose of this retrospective study was to elucidate the clinical characteristics of patients with bilateral pheochromocytomas. We analyze the clinical data of 25 patients with bilateral pheochromocytomas who were treated at Peking Union Medical College Hospital between 1952 and 2004. The average age at diagnosis was (32 +/- 14) years. 19 cases (76%) were familiar type, and among the 19 cases, 13 cases were multiple endocrine neoplasia (MEN), 5 cases von Hippel-Lindau (vHL) disease and 1 case of isolated familial pheochromocytomas. In the 25 patients, bilateral pheochromocytomas were discovered at the same time in 88%, and multiple tumors existed in at least one side of the adrenal gland in 56%. 50% of cases recurred after resection of pheochromocytomas.     The present study was to investigate the mRNA, protein expression and the activity of calcineurin in the hypertrophic heart, and to determine the effect of calcineurin inhibitor--cyclosporine A (CsA) on the regression of cardiac hypertrophy in renovascular hypertensive rats. Renovascular hypertension was induced by two kidney-one clip methods.     To investigate the effects on myocardial injury and sarcoplasmic reticulum (SR) Ca(2+)-ATPase of viral myocarditis mice treated with Astragaloside (AS) and Astragalus Injection (AI). Viral myocarditis model was created by intraperitoneal inoculation with coxsackievirus B3m (CVB3m) solution and were divided into model, AS, AI and normal control groups. The mortality, myocardial pathological changes, serum cardiac troponin I (cTnI) and the activity of myocardial Sarco/Endoplasmic Ca(2+)-ATPase (SERCA) were observed. The mortality of model was higher than that of the normal control (P = 0.0042), AS and AI (P < 0.05).     The mrp (multidrug resistance protein) gene has been associated with the multidrug resistance of cancer cells in vitro and in vivo. To gain information on its physiological role, embryonic stem cells were used to generate mice homozygous for a disruption of the mrp gene, resulting in complete abrogation of mrp expression. No physiological abnormalities were observed, at least up to 4 months of age. Viability, fertility, and a range of histological, hematological, and serum-chemical parameters were similar in mrp(+/+) and mrp(-/-) mice. mrp(-/-) mice displayed an increased sensitivity to etoposide phosphate (2-fold) accompanied by greater bone marrow toxicity, whereas the acute toxicity of sodium arsenite was equivalent in mrp(+/+) and mrp(-/-) mice.     Four new cucurbitane-type triterpene sapogenins, compounds 1-4, together with other eight known compounds were isolated from the acid-hydrolyzed fruits extract of Momordica charantia L. Their chemical structures were established by NMR, mass spectrometry and X-ray crystallography. Compounds 1-7 and 9-12 were evaluated for their inhibitory activities toward protein tyrosine phosphatase 1B (PTP1B), a tyrosine phosphatase that has been implicated as a key target for therapy against type II diabetes. Compounds 1, 2, 4, 7 and 9 were shown inhibitory activities of 77%, 62%, 62% 60% and 68% against PTP1B, respectively. All of these tested compounds were exhibited higher PTP1B inhibition activities than that of the Na3VO4, a known PTP1B inhibitor used as positive control in present study. Structure activity relationship (SAR) analysis indicated that the inhibition activity of PTP1B was associated with the presence and number of -OH groups.

    ABSTRACT Aim: Although the National Free Antiretroviral Treatment Program (NFATP) is effectively administered through a community-based treatment model in China, there are some limitations in this model. No-one has reported the outcomes of highly active antiretroviral therapy as part of a hospital-based HIV treatment model in China. Methods: This study was a retrospective analysis of mortality and viral responses in HIV-positive patients taking highly active antiretroviral therapy regimens in a cohort registered in Ditan Hospital, Beijing, China, the largest infectious disease hospital in North China. Results: This study demonstrated low mortality (3.4% of patients died in a median follow-up of 19 months, with a mortality of 1.7 per 100 person-years) from 2005 to 2010 in Ditan Hospital. In an adjusted Cox proportional hazards regression analysis, CD4 cell counts less than 50 cells/l (adjusted hazard ratio of 5.95 [95% CI: 1.3-28.2]) was the strongest risk factor for death, and switching regimens during treatment was associated with lower risk of death. The study also demonstrated 93.4% of patients were adequate viral suppression, 3% were definite virologic failure, 2% were probable virologic failure and 1.3% were indeterminate. In logistic regression analysis, the strongest risk factors for inadequate viral suppression or failure were single, divorced or widowed (adjusted odds ratio: 4.2; 95% CI: 1.2-14.9) and baseline alanine aminotransferase >100 U/l (adjusted odds ratio: 6.2; 95% CI: 1.5-26.1). Conclusion: The hospital-based HIV treatment model appeared to be successful in improving patient survival and should be considered as an important alternative to the community-based treatment models commonly applied in China.

    ABSTRACT In this paper, we describe a feature based informative model to the second track of this year's KDD Cup Challenge. The goal is to discriminate songs rated highly by the user from ones never rated by him/her. The informative model is used to incorporate different kinds of information, such as taxonomy of items, item neighborhoods, user specific features and implicit feedback, into a single model. Additionally, we also adopt ranking oriented SVD and negative sampling to improve prediction accuracy. Our final model achieves an error rate of 3.10% on the test set with a single predictor, which is the best result of single predictors in all the publicized results on this task, even better than many ensemble models.

    Reirradiation is a major therapeutic modality for patients with locally recurrent head and neck carcinoma. Due to normal tissue tolerances, reirradiation using conventional techniques has a narrow therapeutic ratio in the regional recurrence of nasopharyngeal carcinoma (NPC). Pulsed reduced dose-rate radiotherapy (PRDR), which delivers a series of 0.2 Gy pulses separated by 3 min intervals, is a new reirradiation technique. Head and neck carcinoma cells have high levels of epidermal growth factor receptor expression and cetuximab shows a clear benefit to locally advanced head and neck carcinoma.     ABSTRACT Three new supramolecular compounds [H2TPP](1.5)[SW11VO40]center dot 5CH(3)CN center dot 4H(2)O 1, [H2TPP](2)[SW10V2O40]center dot 4CH(3)CN center dot 3H(2)O 2, and [H2TPP][SW12O40]center dot 4H(2)O 3 composed of [H2TPP](2+) cation and vanadium substituted Keggin-type polyoxometalate (POM) anion [SW12-nVnO40]((2+n)-) (n = 0-2) were prepared and characterized. The third-order nonlinear optical (NLO) and optical limiting (OL) properties of resulting hybrids were studied by using an Nd:YAG laser at 532 nm with a pulse duration of tau = 7 ns. The results demonstrate that all of these supramolecular compounds have significant nonlinear reverse saturated absorption, self-defocusing behavior, and good OL performance, especially for compound 1, which has a second hyperpolarizability gamma value of 7.05 x 10(-29) (esu) and exhibits a comparable OL performance with the most well-known OL materials such as In(Pc*)Cl indicating that they are potentially excellent NLO materials. Remarkably, it was also observed that the degree of vanadium substitution in POM anions corresponding to different onset of reduction potential influences the NLO and OL properties of the resulting compounds: third-order NLO and OL properties of compound 2 are inferior to those of compound 1, and are better than those of compound 3. Both their second hyperpolarizability gamma values and the OL performance were observed to be inversely proportional to the HOMO-LUMO gaps of the resulting compounds. The excited charge transfer from porphyrin to POM anions in the supramolecular compounds when exposed to laser irradiation is thought to play key role in the enhancement of NLO and OL response.

    ABSTRACT The satellite communication systems and satellite navigation systems usually require the antenna to provide compact size, wide beam, circular polarization and high gain at low elevation. Based on these needs, a new type of dual-band wide beamwidth quadrifilar helix antenna (QHA) is designed. The available dual-band is achieved by putting two similar antenna structures operated at different frequency bands in an inner and outer coaxial way, and fed by a split coax balun. A 90° self-phased structure is formed through the quadrifilar helix characteristic to obtain a wide beam and good circular polarization performance. Experimental results of the fabricated antenna show that, the measured results are consistent with the simulated ones, and the antenna can provide good wide-beam circular polarization characteristics in both frequency bands.

    Composite films derived from the water-soluble Keplerate-type polyoxometalate (NH4)42[Mo132O372(CH3COO)30(H2O)72]·ca. 300H2O·ca. 10CH3COONH4 (denoted (NH4)42{Mo132}) and chloroform-soluble tetraphenylporphyrin perchlorate [H2TPP](ClO4)2 are successfully fabricated by a layer-by-layer self-assembly method and characterized by UV-vis spectroscopy and X-ray photoelectron spectroscopy (XPS). The structure of the {Mo132} and [H2TPP](2+) in the films remain intact in light of the results of UV-vis spectroscopy and XPS. UV-vis spectra measurements reveal that the amounts of deposition of {Mo132} and [H2TPP](2+) remain constant in every adsorption cycle in the composite films assembly process. Nonlinear optical properties of the composite films have been investigated by using the Z-scan technique at a wavelength of 532 nm and pulse width of 7 ns. The results show that the composite films have notable nonlinear saturated absorption and self-defocusing effects. The combination of {Mo132} with [H2TPP](2+) can result in composite films with remarkably enhanced optical nonlinearities. The interfacial charge transfer induced by laser from porphyrin to POM in the films is thought to play a key role in the enhancement of NLO response. The third-order NLO susceptibility χ((3)) of the composite films increases with the increase of film thickness.

    To study the adherence, proliferation and osteogenesis of mesenchymal stem cells (MSCs) cultured on different HA/ZrO2 composites. The simplex and graded HA/ZrO2 compo-sites were prepared using dry-laid method. The surface topography of the composites was observed by scanning electron microscope (SEM). The MSCs were isolated from rabbits and cultured on experimental groups (simplex HA/ZrO2 composite, graded HA/ZrO2 composite, pure HA or pure ZrO2 coatings respectively) and control group (ordinary culture plate).     The aim of our study was to evaluate the effect of the S3 (spatial subchondral support) humeral plate and to compare the clinical outcomes and complications after ORIF (open reduction and internal fixation) of proximal humeral fractures using either a S3 plate or a PHLP (proximal humeral locking plate). A total of 144 patients with displaced unstable proximal humeral fractures were treated with either a S3 plate or a PHLP. Each patient had a follow-up at least for one year. We retrospectively collected the data and compared the shoulder functional outcome as well as complications of these two methods. During the one-year follow-up, the average Constant scores gradually improved for both groups. Patients treated with use of an S3 plate had better functional results at three and six months (P < 0.05).     Porphyromonas endodontalis lipopolysaccharide (LPS) is considered to be correlated with the progression of bone resorption in periodontal and periapical diseases. Wnt5a has recently been implicated in inflammatory processes, but its role is unclear in P. endodontalis LPS-induced mediator in osteoblasts. Tribbles homolog 3 (TRIB3) encodes a pseudokinase and has been linked to inflammation in certain situations. Here, we found P. endodontalis LPS induced Wnt5a expression in a dose- and time-dependent manner and it also up-regulated translocation, phosphorylation and transcriptional activity of NF-κB in MC3T3-E1 cells. Bay 11-7082 blocked the translocation of NF-κB and Wnt5a expression induced by P. endodontalis LPS.     Hormone replacement therapy (HRT) is widely used to controlling menopausal symptoms and prevent adverse cardiovascular events. However, the benefit and risk of HRT on cardiovascular outcomes remains controversial. We systematically searched the PubMed, EmBase, and Cochrane Central Register of Controlled Trials databases for obtaining relevant literature. All eligible trials reported on the effects of HRT on cardiovascular outcomes. We did a random effects meta-analysis to obtain summary effect estimates for the clinical outcomes with use of relative risks calculated from the raw data of included trials. Of 1903 identified studies, we included 10 trials reporting data on 38908 postmenopausal women. Overall, we noted that estrogen combined with medroxyprogesterone acetate therapy as compared to placebo had no effect on coronary events (RR, 1.07; 95%CI: 0.91-1.26; P = 0.41), myocardial infarction (RR, 1.09; 95%CI: 0.85-1.41; P = 0.48), stroke (RR, 1.21; 95%CI: 1.00-1.46; P = 0.06), cardiac death (RR, 1.19; 95%CI: 0.91-1.56; P = 0.21), total death (RR, 1.06; 95%CI: 0.81-1.39; P = 0.66), and revascularization (RR, 0.95; 95%CI: 0.83-1.08; P = 0.43). In addition, estrogen therapy alone had no effect on coronary events (RR, 0.93; 95%CI: 0.80-1.08; P = 0.33), myocardial infarction (RR, 0.95; 95%CI: 0.78-1.15; P = 0.57), cardiac death (RR, 0.86; 95%CI: 0.65-1.13; P = 0.27), total mortality (RR, 1.02; 95%CI: 0.89-1.18; P = 0.73), and revascularization (RR, 0.77; 95%CI: 0.45-1.31; P = 0.34), but associated with a 27% increased risk for incident stroke (RR, 1.27; 95%CI: 1.06-1.53; P = 0.01). Hormone replacement therapy does not effect on the incidence of coronary events, myocardial infarction, cardiac death, total mortality or revascularization. However, it might contributed an important role on the risk of incident stroke.

    In the glucose measuring technique by surface plasmon resonance, D-galactose/D-glucose binding protein (GGBP) that can specifically adsorb glucose was introduced, and high-precision specific detection of glucose concentration was realized. In the present paper, the GGBP protein was bound on the surface of SPR sensor through thiol coupling method. GGBP binding experiment was carried out on SPR sensor and then glucose concentration experiment was conducted with this sensor. The results indicated that the SPR sensor had good linearity, stability and repeatability in the range of 0.1-10 mg x dL(-1). SPR sensor bound with GGBP would have great potential and vast development prospects.

    Endoplasmic reticular (ER) membrane modifications play an important role in viral RNA replication and virion assembly but little is known about the involvement of ER-membrane remodeling in the infection cycle of fijiviruses in plant cells. The subcellular localization of Rice black-streaked dwarf virus outer capsid P10 was therefore examined using live-cell imaging. P10 fused to eGFP formed vesicular structures associated with ER membranes in Nicotiana benthamiana epidermal cells and in rice protoplasts. Subcellular fractionation experiments confirmed that P10 is an integral membrane protein. Three predicted transmembrane domains and two less-well-defined domains were each able to target eGFP to the ER.
