    One of the most economically viable processes for the bioconversion of many types of lignocellulosic wastes is represented by edible mushroom cultivation. Pleurotus florida is one of the prominent economically cultivated mushrooms with varied abilities to utilize lignocellulose as a growth substrate. Thus, the profile of oxidative and hydrolytic enzymes was measured during the submerged and solid-state fermentation at Punjab Agricultural University, Ludhiana, Punjab during the winter (rabi) season of the year 2020–21. The intracellular activity of laccase (10.23 IU/mg), manganese peroxidase (9.98 IU/mg), cellobiohydrolase (5.46 IU/mg) was best obtained in P. florida on the 14th day. The extracellular activity of laccase (15.43 IU/ml), lignin peroxidase (11.21 IU/ml), endoxylanases (9.22 IU/ml)were found to be highest on the 14th day.     Mushroom has gained popularity as prized table delicacy with high nutritive and medicinal value. Ergosterol is a biological precursor (a provitamin) to vitamin D2. The contents of vitamin D2 and sterols in some wild and cultivated mushrooms were determined. Vitamin D2 was determined using an HPLC method, including saponification and semipreparative normalphase HPLC purification Lv et al. [1]. Currently HPLC methods are regularly practiced for the estimation / determination of ergosterol [2]. The original procedure for extraction of ergosterol was worked out by Seitz et al. [3].     The genus Pleurotus comprises a group of edible ligninolytic mushrooms with medicinal properties and important biotechnological and environmental applications. The present study reports phytochemical analysis, antioxidant potential and anti-bacterial activities of extracts of three Pleurotus species: P. florida, P. ostreatus and P. sajor-caju. Cultivation of Pleurotus species was carried out on wheat straw substrate and yield of P. sajor-caju was observed to be maximum. Total proteins, peroxidase enzyme activity and ascorbic acid content were estimated on fresh fruit bodies while radical scavenging activity, total phenol content, flavonoid content and antibacterial activity were measured using extracts from dried and powdered mushrooms. P. florida had highest ascorbic acid content while P. sajor-caju had most radical scavenging potential due to flavonoid content.     Casing is an important step during cultivation of button mushroom. It promotes a shift of vegetative mycelium to form pinheads leading to mature basidiocarps. Therefore, An experiment was planned at Punjab Agricultural University, Ludhiana in which farm yard manure (FYM), biogas slurry (BS), burnt rice husk (BRH), spent compost (SC), coir pith (CP) and sandy soil (SS) were used in six combinations to evaluate their impact on yield of Agaricusbisporus strain U3. Casing mixtures were analyzed for the moisture, water holding capacity (WHC), bulk density, pH, electrical conductivity (EC) and mineral analysis. The moisture content of casing mixtures ranged between 33 to 56.3% with maximum in FYM+CP (1:1) and FYM+BS (1:1) which results in more water holding capacity with 160% and 100% for FYM+CP (1:1) and FYM+BS (1:1) respectively.     Mushrooms are rich source of protein, minerals and antioxidants. Nutritive value of mushrooms differs not only among genus but also among species. Nutritional content of mushrooms are being constantly reported. In present study, emphasis is laid on effect of mutational study on nutritional parameters of mushroom. A total of seven mutants of Calocybe indica obtained through physical and chemical mutagenic treatment were subjected to nutritional evaluation. Five mutants (CMU-5, CMN-9, CMN-11, CMN-2 and CMB-4) indicated higher protein content while ash content was also found more for all the mutants except CMN-9. Tocopherol content was also higher for all the mutants except CMN-3. β-carotene was more from 2 mutants, CMU-2 and CMN-9. Lycopene content was better in CMU-2, CMN-9, CME-2 and CMB-4 while ascorbic acid content for CMU-2 and CMN-3 was better than that of the parent.     Ganoderma lucidum has been widely used as a source of potent nutraceutical products. This study was planned to identify and characterize the role of ergosterol in the developmental process of G. lucidum. Four strains of G. lucidum (GL-I-IV) showed a gradual increase in biomass (from 25.52 to 31.72 g) after 3 weeks of growth in mushroom complete medium broth, with maximum biomass observed for strain GL-III. Upon cultivation of G. lucidum strains on wheat straw supplemented with 5% wheat bran, maximum biological efficiency was recorded for the GL-I strain (31.23%), followed by GL-II (26.73%); the number of fruiting bodies were 927 and 693, each weighing 33.7 and 38.6 g, respectively. The amount of ergosterol in the Ganoderma test strains varied among the strains and at different developmental stages, namely, the vegetative mycelium, spawn run, pinhead, and fruiting body phases. The maximum ergosterol content was produced by the GL-I strain during the vegetative mycelium (4601 p.g/g) and reproductive fruiting body (7009 p.g/g) stages. However, strain GL-IV followed by strain GL-II exhibited maximum ergosterol content in the spawn run stage. The ergosterol content was better for GL-II at the pinhead stage. This report indicates that ergosterol content varies among the test strains. Moreover, it increases with each stage of the cultivation process, that is, from spawn run to pinhead to and fruiting body formation.

    Five Pleurotus hybrid dikaryons, developed through cross-breeding of P. florida PAU-5 (PF-5) and P. sajor-caju PAU-3 (PSC-3) were characterized with respect to textural properties, color, and enzymatic and genetic variability. Texture profile revealed significant differences in springiness, resilience, cohesiveness, and chewiness between all hybrids compared to the parents. Among the hybrid cultures, maximum whiteness was reported in hybrid 37, whereas hybrid 8 had minimum whiteness. Three hybrids (16, 37, 42) showed an increased linear growth rate in relation to PF-5, whereas no hybrid showed a higher growth rate than PSC-3. Maximum endoglucanase and xylanase activity was observed in hybrid 46, whereas minimum activity occurred in hybrid 42. Laccase and protease activity was higher in hybrid 37 and 46, respectively.
