Eight transgenic (Cry1Ac -endotoxin) genotypes and one non-GMO cotton (Gossypium hirsutum L.) variety were used in the investigation. Cry1Ac protein was expressed at various extents in several strains. In a greenhouse environment, sixty days after planting, the cry 1Ac gene's expression peaked. At 60 DAP, the variety Bt.CIM-598 (1.871 g/g) expressed the most toxin of any genotype. Throughout the cropping season, larvae of the Helicoverpa armigera were gathered from every section of the nation that produces cotton. H. armigera larvae that were captured in the field were raised on artificial feeds. There was a negative relationship between the DAP and the amount of Cry1Ac protein (-0.332). The greatest amount of death was seen when first-instar larvae consumed the leaves of 60 DAP plants. It was found that transgenic variants had a considerable effect on the percentage mortality of H. armigera. MNH-886 larvae showed the highest mortality percentage when fed on leaves that were har...

Gossypium arboreum, a cultivated cotton species (2n = 26, AA) native to Asia, possesses invaluable characteristics unavailable in the tetraploid cultivated cotton gene pool, such as resistance to pests and diseases and tolerance to abiotic stresses. However, it is quite diffi-cult to transfer favorable traits into Upland cotton through conventional methods due to the cross-incompatibility ofG. hirsutum (2n = 52, AADD) andG. arboreum. Here, we improved an embryo rescue technique to overcome the cross-incompatibility between these two pa-rents for transferring favorable genes fromG. arboreum intoG. hirsutum. Our results indi-cate that MSB2K supplemented with 0.5 mgl-1 kinetin and 250 mg-1 casein hydrolysate is an efficient initial medium for rescuing early (3 d after pollination) hybrid embryos. Eight pu-tative hybrids were successfully obtained, which were further verified and characterized by cytology, molecular markers and morphological analysis. The putative hybrids were subse-que...

Cotton is an important agro-industrial crop across the globe. Improving the fiber quality and yield potential of cotton are major commercial targets for cotton breeders. The cotton lint yield is computed by multiplying three fundamental yield constituents: average boll weight, boll number per unit ground area, and lint percentage. The cotton species Gossypium arboreum exhibits a wide range of desirable traits, which are absent in the congener Gossypium hirsutum. Four parental lines of G. hirsutum and G. arboreum, with significant differences in boll-related traits, were used to develop the following four F2 populations: Mei Zhongmian × Chimu Heizi (MC), Mei Zhongmian × L-02292-3 (ML), Dixie king × Suyuan 04-44 (DS), and Dixie king × Pamuk (DP), in order to study complex traits, such as boll weight (BW) (g), lint percentage (LP) (%), boll upper width (BUW), boll medium width (BMW), boll lower width (BLU), and boll length (BL) (mm). In segregation populations, extensive phenotypic dif...

Combining ability and heterosis for within-boll yield components and fibre quality traits were carried out using line × tester mating system in upland cotton at the University of Agriculture, Faisalabad, Pakistan.Variance due to specific combining ability was greater than general combining ability for all the traits depicting the importance of non-additive genetic effects. The line BH-89 was found to be good general combiner for lint mass per boll, seed volume per 100 seeds, surface area per seed and lint percentage. NIAB-999 was good general combiner for fibre fineness. The third line which was actually the F1 of the first two lines performed better regardingseed number per boll, seed mass per bolland fibre strength.Among testers, NIAB-78 exhibited higher GCA effects for seed number per boll, lint mass per boll and fibre fineness while CIM-1100 proved to be good general combiner for seed volume per 100-seeds, surface area per seed and lint percentage. NIAB-999 × VH-144 exhibited the...

Abstract: Heading date in rice is one of the most important indices in rice breeding being governed by multiple genes known as quantitative trait loci (QTLs). This character along with grain yield determines the commercial potential of any rice cultivar. In the last three decades the development of DNA markers have led to the precise analysis of QTLs. Various mapping populations are being used to explore QTLs for yield related parameters including heading date in rice. Like F 2, RILs, NILs, DH, BCs and SSSLs. To date among reported QTLs of heading date, 20 QTLs have been identified and cloned. The purpose of the present review is therefore to consolidate our existing knowledge about the progress that so far have been made in mapping of QTLs for heading date character. In addition to that requirements of QTL mapping and different QTL mapping methods have also been discussed. The information could be useful for further identification of QTLs for heading date that can be successfully i...

One hundred spring wheat accessions were assessed for heat tolerance under plastic sheet tunnel resulting in seven parents with diverse heat tolerance. Tolerant and susceptible genotypes were graded on the bases of their relative cell injury percentages and relative values for different yield components. The analysis of variance for relative cell injury % revealed highly significant differences among the genotypes with a range from 28 to 98 %. These 7 parents were crossed in a full diallel system to evaluate the inheritance pattern of some spike related yield attributes (spike length, spikelets per spike, spike density, spike weight, grains per spike and grain yield per plant) under different temperature regimes at grain filling stage. Preliminary ANOVA revealed significant genotypic variation (P<0.01) for all the traits studied under both environments. Spike length, spikelets per spike and spike density revealed partial fitness of data for additive dominance model under normal c...

The ability of plant to sustain itself in limited water conditions is crucial in the world of agriculture. To breed for drought tolerance in wheat, it is essential to clearly understand drought tolerant mechanisms. Conventional breeding is time consuming and labor intensive being inefficient with low heritability traits like drought tolerance. Recent progress made in the field of genomics enabling us to access genes linked with drought tolerance has enhanced our understanding of this complex phenomenon. The purpose of this review paper was to briefly overview the accomplishments in molecular breeding for drought tolerance in wheat. Thus, by knowing the genetics of drought tolerance and identifying quantitative trait loci (QTLs) linked with DNA markers will help wheat breeders to develop high yielding drought tolerant cultivars. Key words: Triticum aestivum L, drought tolerance, QTLs, marker assisted selection (MAS).

The present study was undertaken for the estimation of heritability, genetic variability and genetic advance of yield contributing traits in F 3 generation of upland cotton (Gossypiumhirsutum L.). The F 3 generations of ten crosses along with their parents were sown in non-replicated field trail. The data was recorded on maturity for node of first fruiting branch, total number of nodes/plant, plant height (cm), monopodial branches per plant, sympodial branches per plant, total number of fruiting points, number of bolls/plant, boll retention percentage, average boll weight, yield per plant (kg), lint percentage and fiber length (mm). The heritability in broad-sense, genetic advance and genetic variability estimates presented in table-3, table-4 and table-5 suggested that the highest heritability in broad–sense coupled with good genetic advance and genetic variability was present for plant height (83.78%, 10.51 and 39.75), boll retention percentage (81.78%, 4.54 and 8.18), fiber lengt...

A study was conducted in the Department of Plant Breeding and Genetics, University of Agriculture Faisalabad, Pakistan during 2005-06. In this study five wheat genotypes viz. Chenab-2000, Rohtas-90, SH-2002, Uqab-2000 and 243-1 were crossed in complete diallel fashion to study gene action for plant height, peduncle length, flag leaf area, number of tillers per plant, 100-grain weight and grain yield per plant. Analysis of variance proved to be significant for all characters. The significant deviation of joint regression value “b” from zero and non-significance of “t” square value, suggested the absence of epistasis for all traits which in turn attested fitness of the data for simple additive dominance model. Additive genetic component “D” proved to be significant for all traits, but dominant components H1 and H2 were higher in magnitude for plant height, flag leaf area, number of tillers per plant and grain yield per plant. It illustrated the preponderance of dominance genetic effec...

Cotton is the backbone of Pakistan’s economy. More than half of the export of Pakistan depends on cotton and cotton-based products. Morphological quantitative traits are important for estimation of cotton yield. In this regard, a study was conducted to evaluate the morphological responses on cotton crop at experimental farm of MNS-University of Agricultural Multan during the year 2019-2020. Four cotton varieties with different time windows (early, normal and late) were sown in a randomized complete block design (RCBD) in a split plot arrangement. Correlation and regression studies of cultivars of Gossypium and Malvaceae were analyzed for quantitative characters. Result revealed that significantly positive correlation of boll weight (r=0.211), seed volume (r=0.138), seed oil content (r=0.020) and negatively correlation with seed index (r= -0.005) was displayed by all traits with seed cotton yield, which showed that seed cotton yield was greatly influenced by all characters. Regressio...

Cotton is a major cash crop and backbone of the textile industry in Pakistan which is badly affected by sucking insects. Drought is an important abiotic factor in trichome development. The objective of the study was to determine the effects of drought on trichome density and length. Trichome density was measured in two ways, one through the scaling method and the other through counting the trichome density manually. The scaling method is qualitative grading while quantitative grading includes trichomecount in a card of optimized length. Three scales were finalized to classify leaves on the basis of trichomes which were counted in a specific area (0.25cm2) on abaxial side of the leaf. In drought stress, trichomes density and length were measured and compared to that in normal conditions. Trichome density varies from 12 to 56 in 0.25cm2 under drought stress. On the basis of correlation of trichome density with stomatal conductance, photosynthetic rate, PAR and transpiration ratio unde...

BACKGROUND Cotton is the world's richest source of natural fiber. Meanwhile cotton plant is top ranked stress sensitive plant thereby affecting its yield and fiber quality. But, in climate change scenario, fiber yield and quality are being affected due to environmental stresses, especially heat, drought and salinity. Present study is aimed to identify cotton genotype harboring prominently expressed stress responsive genes. METHODS Four cotton genotypes (IUB-13, IUB-222, IUB-09 and MM-58) were evaluated under drought and salinity stress for yield traits and expression of different stress responsive genes (GhWRKY3, GhDREB2 and GhRDR6). RESULTS Pronounced expression of GhWRKY3, GhDREB2and GhRDR6 was observed in cotton variety IUB-13 in stress condition (drought and salinity) as compared to control followed by IUB-222 which revealed that these genotypes might possess substantial potential to cope with environmental hazards encountered in growing season CONCLUSION: Utilization of cotton genotypes i.e., IUB-13 and IUB-222 in cotton breeding program can be very much fruitful for developing cotton genotypes adoptable to climate change.

Introduction. The inheritance studies on hypoxia tolerance in cotton are very scanty. Objective. The objective of this work was to study the inheritance pattern of seed cotton yield, ginning out-turn, and fiber length under normal and hypoxia conditions. Materials and Methods. An 8•8 diallel mating system was employed in F1 generation of cotton. The experiment was conducted during the years 2011-2012 at Cotton Research Institute, Multan, Pakistan. Results. Analysis of variance revealed significant variation (P<0.01) for all the traits under both non-stress and flooding stress conditions. The value of regression coefficient b deviated significantly from zero but not from unity for all the traits which enabled further analysis of the data. The additive component (D) was significant for all the traits and this has more values than the values of dominance components H1 and H2, which clearly indicates the preponderance of additive genetic effects. Under normal and hypoxia conditions t...

This study was conducted to determine the association between various drought tolerance traits with yield, genetic diversity for drought tolerance among various Cotton genotypes and mapping of Quantitative trait loci (QTLs) conferring drought tolerance in Gossypium hirsutum L. Six genotypes (CIM-557, CIM-573, Cyto-108, Cyto-109, BH-172, CRIS-121) having significant morphological and physiological differences for drought tolerance were selected. BH-172 performed well under drought stress while Cyto-109 showed meager performance under similar conditions. A positive correlation was observed between yield and boll per plant, boll weight, height of the main stem, internodal length, stomatal conductance, photosynthetic rate, leaf area index, and leaf relative water potential while, node on the main stem, transpiration rate, canopy temperature, chlorophyll content and proline contents showed a negative correlation with seed cotton yield. Dendogram and similarity matrix assigned cotton cult...
