The aim of this study was to investigate the correlation between biochemical and molecular markers in Eucalyptus microtheca F. Muell. under in vitro culture. For this mean, twig-derived explants obtained from Eucalyptus microtheca 1-year-old seedling were cultured on modified MS medium, supplemented with different concentrations of NAA, Kin and TDZ. POD (peroxidase) alleles patterns were studied among regenerated plantlets to investigate the effect of TDZ concentration on POD activity. A dimer locus, a tetramer locus and two epigenetic bands were observed. Genome variation among somaclonal plantlets were investigated using microsatellite markers. SSR (Simple Sequence Repeat) markers revealed polymorphism among the studied population. Nonparametric statistical analysis showed significant effect of simple sequence repeats loci on peroxidase alleles. Correlation of two similarity matrix POD and SSRs loci was 0.18 using Mental test. Results showed less stability of dimer locus in different concentrations of TDZ compared to tetramer locus. Tetramer alleles showed more correlation to SSRs than that of dimmer ones.

Objective: The present study was done to investigation the optimal conditions for hazelnut plants regenerated from cotyledon as explants and invitrotaxol production. Methods: Cotyledon segments were cultured on MS medium containing 2,4-D concentrations of 0, 0.5 and 1 mgl-1 alone or in combination with concentrations of 0, 0.5 and mgl-1 BA for callus induction. For regeneration, Calli were sub-cultured to the same medium and or transferred to the MS medium containing BA at concentrations of 0, 0.25, 0.5, 1, 2 and 3 mgl-1 Callus was formed in all hormonal treatments. In order to investigation of genetic variation among lentil lines and genetic relationship these to Cistan local cultivar, DNA of 20 lentil lines and Cistan local cultivar were analysed using random amplified polymorphic DNA (RAPD) and intron-exon splice junctions (ISJ) markers. PIC, MI, percentage of polymorphic bands (P), Nei , s gene diversity (H), Shannon , s information index (I) were calculated for both random, semi random and combined of random and semi random primers. For both matrices, genetic similarity (GS RAPD and GS ISJ ) were calculated according to the Jaccard similarity coefficient. Correlation between two similarity matrices obtained with two markers types was estimated. The similarity matrix per marker was subjected to cluster analysis by the unweighted pair-group method (UPGMA).
