DATE:

TITLE: Enzymes
AIM: TO OBSERVE THE ACTION OF THE ENZYME CATALASE
MATERIALS/APPARATUS: one potato, one scalpel, one ruler, four test tubes, one 250 ml
beaker, one tripod, one Bunsen burner, one wooden splint, one mortar and pestle, Hydrogen
Peroxide, Hydrochloric acid, one 10 ml syringe and one pair of forceps.
METHOD/PROCEDURE:
(1) Using the scalpel and ruler four pieces of potato were cut into 1 cm³ cubes.
(2) Using the 10 ml syringe, 2 ml of Hydrogen peroxide was placed into a test tube.
(3) Forceps were then used to place one potato cube into the test tube and observations were
made and recorded.
(4) Using the 250 ml beaker, tripod and Bunsen burner, a water bath was set up.
(5) Forceps were used to place another potato cube into the water bath and this was left for
two minutes.
(6) After the time had elapsed the potato cube was removed from the water bath and placed
into another test tube filled with 2 ml Hydrogen peroxide. Observations were made and
recorded.
(7) The third potato cube was crushed using the mortar and pestle and placed into another
test tube containing 2 ml Hydrogen peroxide. A glowing splint was held over the mouth
of this test tube and observations were made and recorded.
(8) Using the 10 ml syringe, 2 ml of Hydrogen peroxide was placed into a test tube followed
by 1 ml of Hydrochloric acid. The last potato cube was then added to this test tube and
observations were made and recorded.

DISCUSSION:

° What is an Enzyme?
- reactions occurring in living organisms without being changed themselves.

° Why are Enzymes important to living organisms?

° Discuss the factors that affect the rate of enzyme activity

° What is Catalase and where is it found?

° Describe and compare the amount of bubbles formed when the enzyme was exposed to
different conditions.

° Discuss why the activity occurred the way it did under each condition.

° Compare your results with what was expected to occur in your theory (factors that affect
enzyme activity)

° Why did the glowing splint relight?

° How could the experiment be done better?

PRECAUTIONS:
° Gloves were used when handling the Hydrogen Peroxide and Hydrochloric acid as both can
cause irritations of the skin.

SOURCES OF ERROR:
° Hydrogen Peroxide decomposes when exposed to light (it is usually kept in a dark bottle).
Some decomposition may have occurred when the H202 was placed into the test tubes.

° The different potato cubes may have contained varying concentrations of the enzyme Catalase.

CONCLUSION:
From the experiment it can be concluded that the action of the enzyme Catalase is affected by
Temperature, Surface Area and pH.
