Directions:
1. Collect the water sample in the container provided
2. Open the pack. Remove one plate from the kit
3. With the help of provided syringe, inject 0.1 ml to 0.5ml of water sample into the
plate.
4. The sample will be evenly absorbed.
5. Incubate the plate at room temperature (28-30°C) for 24 hours.
6. Read the plate and count the colonies.

Principle and Interpretation:
Vibrio species are gram negative bacteria. They are salt tolerant and are found in environments with higher salinity.
Vibrio has a major occurrence in sea food, which can lead to food poisoning. This gives Vibrio clinical significance.
Regular monitoring of samples for presence of Vibrio can prevent outbreaks and health hazards.
Detection of Vibrio species include enrichment in Alkaline Peptone water and further plating on selective media.
The most commonly used selective medium includes TCBS Agar based on sucrose fermentation with
bromothymol blue and thymol blue as indicator. Bile and high pH add to the selectivity of the medium. Other
most common medium used for enumeration and isolation include selective and differential medium like CPC
Agar base, Vibrio Agar, Proteose Agar, VP Medium, Vibrio Parahaemolyticus Sucrose Agar, Wagatsuma Agar
Base.
Chromogenic medium for Vibrio is more specific for Vibrio species wherein Vibrio cholerae gives purple colour,
Vibrio parahaemolyticus gives green colour and Vibrio fluvialis gives blue colour. The medium is selective and
inhibits the unwanted accompanying microflora.
