Acid soluble collagen (ASC) was extracted from the skins of young and adult Nile perch (Lates niloticus) using 0.5 M acetic acid and precipitation with 0.9 M NaCl. The ASC yields, on a dry weight basis, were 63.1 and 58.7%, respectively for young and adult fish skins. SDS-PAGE showed that the collagens contained two alpha components (α1 and α2). ASC from Nile perch was found to contain more imino acids (19.3 and 20.0%, respectively, for young and adult fish) than most fish species. The denaturation temperature for the collagens from the skins of young and adult Nile perch was determined to be 36 °C, which is also higher than that for most other fish species. Fourier transform infrared spectroscopy showed a higher degree of molecular order in ASC from adult than from young Nile perch. The results indicate that age-related changes in Nile perch skin collagen are not very pronounced, probably because there is minimal development of mature cross-links.

The influence of NaCl salinity on growth, dry-matter production and leaf photosynthesis of seedlings of Eucalyptus camaldulensis Dehnh. and Dalbergia sissoo Roxb. was studied by imposing 4 levels (40, 80, 120 and 160 mM) of NaCl in pot culture. Salinity up to 160 mM did not affect plant survival, but did affect plant growth and dry-matter production depending upon the species and salt concentration. NaCl reduced leaf number and dry-weight of all the plant components, but increased stem dry-weight, especially in E. camaldulensis. Salinization also stimulated total dry-matter production at all the salinity levels in E. camaldulensis but only at 40 mM in D. sissoo. The two species varied in protein and chlorophyll concentration and in leaf photosynthetic rate. Protein and chlorophyll concentration of the plants fell at all the levels of NaCl, except at 40 mM, where stimulation in the photosynthetic carbon assimilation of the plants occurred. However, no distinct relationship between leaf photosynthetic rate and dry-matter production was found. The study indicated that low salt concentrations generally stimulated growth, biomass production and rate of photosynthesis in both the species, and E. camaldulensis appeared more NaCl salt-tolerant than D. sissoo.

Sediment traps were deployed on the Northwest Shelf (NWS) of Australia in November 1996, to determine fluxes of organic matter and inorganic elements from the photic zone to deeper waters in a transect extending from Exmouth Shelf to Exmouth Plateau. Infiltrex II water samplers collected particulate and dissolved organics from the water column near the trap sites. Surface sediments and sediment cores were also collected over the study region. Lipid biomarkers were used to determine the sources of organic carbon and its cycling processes on the NWS.Dry weight fluxes from the traps ranged from 124 to 616 mg m−2 day−1and particulate organic carbon (POC) fluxes ranged from 22 to 42 mg m−2 day−1. The biogenic lipids consisted of biomarkers indicative of marine zooplankton, phytoplankton and bacteria, plus traces of land plant markers. A large contribution of unresolved complex material (UCM), which is indicative of petroleum, was detected at four times the biogenic hydrocarbon flux at shallow stations, and up to seven times the biogenic hydrocarbon flux at the most offshore station.There is essentially no river input, and only trace aeolian-derived material to contribute to primary production on the NWS of Australia. Most of the organic matter produced are rapidly recycled in the water column and the small fraction of lipids that settle to the sediments is already partially degraded and undergoes further rapid degradation in the surface sediments. Natural oil seeps also provide utilisable organic carbon to the system. The production and vertical flux rates of organics determined in this study are comparable to those reported in studies of shallow traps in oceanic areas from long-term studies in the Arabian Sea, and other coastal margins such as the Bay of Biscay (France) and California (USA).In offshore areas, most “living” lipid materials passed through the GFF filters thus invalidating POC estimates based on high volume sampling. To adequately assess living (particulate) carbon, gentle filtration of low volume seawater samples is more accurate, as shown by this solid phase absorption study of individual lipid biomarkers.

The relationship between dry weight and body length for larvae of Plecoptera (Leuctra spp., Isoperla grammatica, Nemoura cinerea) and Ephemeroptera (Baetis spp., Habrophlebia fusca, Paraleptophlebia submarginata, Ecdyonurus helveticus, Rhithrogena semicolorata), collected from a carbonate stream in the Apennine (central Italy), is reported. The power equation f(x) = Ax B has been applied to fit the curves of dry weight vs. body size (length) in the ranges 0.03–13.00 mg and 2–14 mm, respectively; a total of 674 larvae were examined. The power model was in very good agreement with experimental data. Moreover, the error between measured and estimated weight was in the 4–20% range. The data on Isoperla grammatica, Leuctra spp., Rhithrogena semicolorata and Baetis spp. were compared to those in a previous study in a different geographical setting (south-western Germany’s Black Forest) obtaining similar results but with lower errors. We used and compared two methods: the weighted least-square method (WLS) and an analysis of covariance (ANCOVA). The values of the A and B coefficients obtained with the two methods were very similar (A or B). We found the best fits for all the examined Plecoptera (species, genus, and order level), while the results for Ephemeroptera were varied, with loose fits at the order level and also for Leptophlebiidae collectively considered.

The present work describes a coke oven soil treatability study by landfarming, conducted on-site in a field scale facility covering 100 m2. The soil contamination was mainly due to high concentrations of polynuclear aromatic hydrocarbons (PAHs) up to 1140 mg/Kg dry weight (ΣEPA). Along the treatment process the soil was characterised at the chemical, microbiological and ecotoxicological levels. After 3 months a reduction of 63% in total PAHs concentration was observed, being detected a more pronounced reduction for PAHs with 2, 3 and 4 rings (79%). Concomitantly, a change in the composition of the microbial population was observed with a significant increase in the PAHs degrading and total heterotrophic colonies. Concerning the ecotoxicity and genotoxicity data no effect was detected in the treated soil samples eluates.
