Microbial fuel cells (MFCs) are biochemical-catalyzed systems in which electricity is produced by oxidizing biodegradable organic matters in presence of either bacteria or enzyme. This system can serve as a device for generating clean energy and, also wastewater treatment unit. In this paper, production of bioelectricity in MFC in batch and continuous systems were investigated. A dual chambered air–cathode MFC was fabricated for this purpose. Graphite plates were used as electrodes and glucose as a substrate with initial concentration of 30 g l−1 was used. Cubic MFC reactor was fabricated and inoculated with Saccharomyces cerevisiae PTCC 5269 as active biocatalyst. Neutral red with concentration of 200 μmol l−1 was selected as electron shuttle in anaerobic anode chamber. In order to enhance the performance of MFC, potassium permanganate at 400 μmol l−1 concentration as oxidizer was used. The performance of MFC was analyzed by the measurement of polarization curve and cyclic volatmmetric data as well. Closed circuit voltage was obtained using a 1 kΩ resistance. The voltage at steady-state condition was 440 mV and it was stable for the entire operation time. In a continuous system, the effect of hydraulic retention time (HRT) on performance of MFC was examined. The optimum HRT was found to be around 7 h. Maximum produced power and current density at optimum HRT were 1210 mA m−2 and 283 mW m−2, respectively.► Power generation was investigated in a single MFC at batch and continuous modes. ► Continuous MFCs offer some advantages over batch systems for practical applications. ► Polarity and cyclic voltammetry, were adopted to analyze experimental data. ► OCV was stable for the duration of 72 h of operation time in batch system. ► At optimum HRT (6.7 h), maximum output were 1210 mA m−2 and 283 mW m−2, respectively.

Laccase from the white rot fungus Coriolopsis polyzona was immobilized for the first time through the formation of cross-linked enzyme aggregates (CLEAs). Laccase CLEAs were produced by using 1000g of polyethylene glycol per liter of enzyme solution as precipitant and 200muM of glutaraldehyde as a cross-linking agent. These CLEAs had a laccase activity of 148Ug(-1) and an activity recovery of 60.2% when using 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) as substrate. CLEAs formed by co-aggregation with bovine serum albumin (BSA) as a stabilizer showed lower laccase activity and affinity for ABTS than those without BSA. The CLEAs co-aggregated with BSA showed higher residual activity against a protease, NaN(3), EDTA, methanol and acetone. Earthworms’ body works as a ‘biofilter’ and they have been found to remove the 5 days’ BOD (BOD5) by over 90%, COD by 80–90%, total dissolved solids (TDS) by 90–92%, and the total suspended solids (TSS) by 90–95% from wastewater by the general mechanism of ‘ingestion’ and biodegradation of organic wastes, heavy metals, and solids from wastewater and also by their ‘absorption’ through body walls. Earthworms increase the hydraulic conductivity and natural aeration by granulating the clay particles. They also grind the silt and sand particles, increasing the total specific surface area, which enhances the ability to ‘adsorb’ the organics and inorganic from the wastewater. Intensification of soil processes and aeration by the earthworms enable the soil stabilization and filtration system to become effective and smaller in size. Suspended solids are trapped on top of the vermifilter and processed by earthworms and fed to the soil microbes immobilized in the vermifilter. There is no sludge formation in the process which requires additional expenditure on landfill disposal. This is also an odor-free process and the resulting vermifiltered water is clean and disinfected enough to be reused for farm irrigation and in parks and gardens
