Obesity is associated with inflammation and immune cell recruitment to adipose tissue, muscle and intima of atherosclerotic blood vessels. Obesity and hyperlipidemia are also associated with tissue insulin resistance and can compromise insulin delivery to muscle. WhenMortierella alpina ATCC 32222 was incubated in a glucose salts medium at 25°C the biomass (17.5 g/l) contained 9.62% arachidonic acid which amounted to 54% (w/w) of total biomass lipids. When the glucose concentration in the medium was varied from 0 to 150 g/l, the percentage of arachidonic acid in biomass and in lipids was highest at a glucose concentration of 30 g/l, but highest yield of arachidonic acid per litre of culture broth was observed at a glucose concentration of 100 g/l. While production of biomass reached a plateau of 17 g/l after a 3-day incubation at 25°C, the percentage of arachidonic acid in lipids and biomass increased dramatically from 3 to 6 days with a concurrent arachidonic acid yield increase from 0.89 to 1.63 g/l. Optimum initial culture pH for arachidonic acid production was in the range 6.0–6.7. By increasing the concentration of the glucose salts medium three-fold, yields of biomass and arachidonic acid were increased to 35.8 g/l and 3.73 g/l, respectively.

Ten margarine brands of Pakistan were analyzed for their fatty acid composition with emphasis on trans fatty acids (TFA) using GC-MS. Saturated, cis-monounsaturated and polyunsaturated fatty acids were present at 24.2–58.1, 5.7–35.4 and 3.8–37.4% of total fatty acids, respectively. Among the saturated fatty acids, palmitic acid (16.9–33.8%) was dominant in all analyzed margarine brands and its higher amount indicates that palm oil was a major contributor in the margarine manufacturing. Among samples tested only one contained a low level of TFA (2.2%) while the rest contained very high amounts of TFA (11.5–34.8%) which clearly shows that hydrogenated oils were used in the formulation of margarines. Fatty acid profiles demonstrated that all samples belong to the hard margarine category containing high amounts of trans and saturated fatty acids which is an alarming issue for the health of consumers.

Commerical biodiesel is composed of FAME. It may also contain small amounts of FA, which are quantified by an acid number, expressed as milligrams of potassium hydroxide required to neutralize 1 g of sample. In 2006, the ASTM D 6751 biodiesel acid-number limit was harmonized with the European biodiesel value of 0.50. ASTM D 664 is the standard reference method for measuring the acid number of both ASTM biodiesel and petroleum-derived diesel. This potentiometric method cites acceptable repeatability and mediocre reproducibility, but no information on accuracy. ASTM D 974 is a non-aqueous colorimetric titration that uses potassium hydroxide in isopropanol as the titrant and p-naphtholbenzein as indicator. It was designed for petroleum products and is suitable for colored samples. It has been tested on nine palmitic acid/soybean oil standards in the acid-number range of 0.198 to 1.17. All accuracies were within 3.3%. The repeatability was approximately 6% at an acid number of 0.5. The reproducibility appears to be only slightly greater than the repeatability at an acid number of 0.5. It is concluded that ASTM D 974 is a good method for evaluating the acid-number compliance of biodiesel samples.

The physico-chemical characteristics of the seeds and seed oils of four citrus species, Mitha (Citrus limetta), Grapefruit (Citrus paradisi), Mussami (Citrus sinensis), and Kinnow (Citrus reticulata) were investigated. The hexane-extracted oil content of citrus seeds ranged from 27.0 to 36.5%. The protein, fiber and ash contents were found to be 3.9–9.6%, 5.0–8.5%, and 4.6–5.6%, respectively. The extracted oils exhibited an iodine value of 99.9–110.0; refractive index (40 °C), 1.4639–1.4670; density (24 °C), 0.920–0.941 mg/mL; saponification value, 180.9–198.9; unsaponifiable matter, 0.3–0.5%; acid value (mg KOH/g of oil), 0.5–2.2 and color (1-in. cell) 1.4–3.0R + 15.0–30.0Y. The oils revealed a good oxidative stability as indicated by the determinations of specific extinctions at 232 and 270 nm (2.3–4.4 and 0.6–0.9, respectively), p-anisidine value (2.2–3.2) and peroxide value (1.6–2.4 mequiv/kg of oil). The citrus seed oils mainly consisted of linoleic acid (36.1–39.8%). Other prominent fatty acids were palmitic acid (25.8–32.2%), oleic acid (21.9–24.1%), linolenic acid (3.4–4.4%), and stearic acid (2.8–4.4%). The contents of tocopherols (α, γ, and δ) in the oil were 26.4–557.8, 27.7–84.1, and 9.1–20.0 mg/kg, respectively. The results of the present study demonstrated that the seeds of citrus species investigated are a potential source of valuable oil which might be utilized for edible and other industrial applications.

Seed oils from six legume cultivars of Phaseolus vulgaris, grown in the Kingdom of Lesotho, were extracted and their physicochemical properties and FA compositions were determined in order to compare their dietary lipids with those in P. vulgaris cultivars grown in other parts of the world. The oil content of the beans was very low, ranging from 1.5 to 2.0% (w/w). The acid values ranged from 11.0 to 19.2 mg KOH/g, whereas a combination of the PV and the p-anisidine values in Holm's equation gave oxidation values that ranged from 11.0 to 15.0. Thus, considerable enzymatic hydrolysis and oxidation had taken place in the beans during storage. Iodine values ranged from 80.5 to 92.3 (Wijs method), indicating moderate unsaturation in the oils. However, capillary GC analysis, supported by proton NMR analysis of the FAME, gave a total unsaturation range from 79.67 to 84.24%. The dominant FA were α-linolenic acid (36.47–48.81%) and linoleic acid (20.96–36.10%), with appreciable amounts of palmitic acid (14.33–18.23%). This FA composition pattern is quite similar to the FA distribution reported for low oil-bearing legume seeds. Thus, notwithstanding the different climatic and soil conditions, the general properties of lipids in the southern African legume cultivars were quite similar to those of lipids in P. vulgaris cultivars grown in other parts of the world. The high content of α-linolenic acid in the cultivars of P. vulgaris could very likely play a beneficial role in reducing the risk of coronary heart disease among the large populations consuming them in the southern African region.

The oil yield, fatty acid composition and the physicochemical and quality characteristics of chia crude seed oils obtained by pressing and solvent extraction were determined. The extraction methods assayed influenced significantly the oil yield, obtaining about 30% more oil by solvent than by pressing. The main fatty acids ranked in the following order of abundance: α-linolenic acid (α Ln) > linoleic acid (L) > oleic acid (O) ≈ palmitic acid (P) > stearic acid (S) for both extraction systems. The n-3/n-6 FA ratio of chia oils ranged from 3.18 to 4.18, being markedly higher than that reported for other vegetable oils. The main triacylglycerols were: αLnαLnαLn > αLnαLnL > αLnLL > αLnαLnP > αLnLO ∼ αLnLP, which represent about 87–95% of the total content of these compounds. The quality and composition of some minor constituents of chia seed oils were influenced by the extraction process. Oils presented a moderate content of bioactive components, such as tocopherols, polyphenols, carotenoids and phospholipids; the high unsaturation level determined their low oxidative stability.

Nutritional diets are essential to prevent nonalcoholic fatty liver disease (NAFLD) in the global obesity and diabetes epidemic. The ingestion of palmitic acid-rich diets induces NAFLD in animal and human studies. The beneficial properties of olive oil (oleic acid) may be superseded by ingestion of palmitic acid-rich diets. Hepatic caffeine metabolism is regulated by palmitic and oleic acid with effects of these fats on amyloid beta metabolism. Healthy fats such as olive oil may facilitate rapid amyloid beta clearance in the periphery to maintain drug therapy in diabetes and various neurological diseases. Repression of the anti-aging gene sirtuin 1 (Sirt 1) prevents the beneficial properties of olive oil. Brain disorders induce NAFLD and supersede caffeine's therapeutic effects in the prevention of NAFLD. Delayed hepatic caffeine metabolism in NAFLD and increased caffeine transport to the brain with aging-induced mitophagy in neurons with induction of type 3 diabetes and neurodegenerative disease.

The production of recombinant therapeutic proteins is one of the fastest growing sectors of the pharmaceutical industry, particularly monoclonal antibodies and Fc-fusion proteins. Currently, mammalian cells are the dominant production system for these proteins because they can perform complex post-translational modifications that are often required for efficient secretion, drug efficacy, and stability. These protein modifications include misfolding and aggregation, oxidation of methionine, deamidation of asparagine and glutamine, variable glycosylation, and proteolysis. Such modifications not only pose challenges for accurate and consistent bioprocessing, but also may have consequences for the patient in that incorrect modifications and aggregation can lead to an immune response to the therapeutic protein. This mini-review describes examples analytical and preventative advances in the fields of protein oxidation, deamidation, misfolding and aggregation (glycosylation is covered in other articles in this issue). The feasibility of partially replacing traditional analytical methods such as peptide mapping with high-throughput screens and their use in clone and media selection are evaluated. This review also discusses how further technical advances could improve the manufacturability, potency, and safety of biotherapeutics.

Compositions and concentrations of fatty acids, lipid classes, sterols, fat-soluble vitamins and β-carotene were determined in extracted lipids from prickly pear [Opuntia ficus-indica (L.) Mill] peel. Total lipids (TL) recovered were found to be 36.8 g kg−1 (on dry weight basis). The level of neutral lipids was the highest, followed by glycolipids and phospholipids, respectively. Among the TL, linoleic acid was the dominating fatty acid, while oleic and palmitic acids were estimated to be in relatively equal amounts. Compared with the neutral lipids, the polar fractions were generally characterised by higher percentages of saturated fatty acids and lower percentages of unsaturated fatty acids in all subclasses. Concerning trienes, γ-linolenic acid was present at 8.60% of TL, while α-linolenic acid was present at 0.69%. Recovered lipids were characterised by a high percentage of unsaponifiables (12.8% TL) and found to be a rich source of vitamin E and sterols. Free sterols accounted for ca. 29% of the total unsaponifiables, wherein β-sitosterol and campesterol were the major sterols. In terms of vitamin E, α-tocopherol constitutes about 80.0% of total vitamin E present, the rest being β-, γ- and δ-tocopherols in decreasing order. Moreover, lipids under investigation were characterised by a high levels of β-carotene and vitamin K1. The information obtained in the present investigation is useful for characterising lipid of prickly pear peel and further chemical and nutritional investigations of prickly pear peel. The results are also important for industrial utilisation of the major by-product of the fruit.

NMR-based metabolic profiling was used to assess the effect of Trichostatin A (TSA) treatment on the endogenous metabolome of cultured primary hepatocytes. TSA-treated cultures were metabolically distinct from nontreated cells and the time (44 and 92 h) related effect on metabolism was also reduced. Our data suggest that TSA treatment improves the model for the study of hepatic metabolism and as an in vitro carcinogenicity screening assay.
