For more than 40 years, Shimadzu Corporation has refined the technological capabilities of
high-speed and ultra-high performance liquid chromatograph, and has provided products with
outstanding performance, such as autosamplers with low carryover and detectors which support
stable signal intensity during ambient temperature fluctuations. The new Nexera series aims to
set new standards for high performance liquid chromatography in the world of analysis,
combining a solid foundation of technological know-how with new functions involving M2M,
IoT and AI. A variety of sensors update operating status and instrument configuration to cloud
servers in real time, including data related to consumable parts such as columns and UV lamps,
guaranteeing data integrity and traceability. Sensor data is also used to predict problems such as
mobile phase depletion and wear-and-tear of consumable parts. Pumping issues that can be
caused by air bubbles are automatically detected and resolved via the autopurge function. The
integration of these technologies allows users to generate accurate and reliable chromatographs
without the need for extensive technical knowledge or experience.
New features contribute to more efficient workflow in the lab as a whole. The installation
area required for the LC has been further reduced, and continuous analysis of thousands of
samples has been enabled. It is now possible to collect data from two different analysis paths in
a single system. Basic performance has also been improved thanks to the addition of a UV cut-
off filter to prevent degradation of compounds and three-way temperature control in the optical
system to ensure baseline stability.
These features are shared by all four models in the new Nexera series, which have a
maximum pressure of 130, 105, 70 and 44 MPa respectively, allowing customers with various
analysis needs to experience the benefits of these new technologies.

2. Key features and system configuration


2.1. Key features
The new Nexera Series is an entirely new LC system that represents a new step in the
development of basic HPLC performance, while also incorporating a number of new
features involving the latest digital and information technologies, such as M2M, IoT and
AI.

2.1.1. A New Benchmark of Intelligence


This tagline represents the most notable difference between the new Nexera series and
previous Shimadzu LCs. New features allow the simple management of resources in the
lab and automatic optimization of analysis. Data from various sensors is collected in the

cloud, and thanks to advanced information processing technologies, the status of the
instrument and all consumable parts is recorded during every stage of analysis, and
instrument problems can be forecast and prevented. In the case that an error does occur,
the user can be notified by email, and there are options for the instrument to attempt auto-
recovery without the need for user intervention.
These advances in automation allow for smoother analysis and allow more
inexperienced users to collect reliable data consistently. The system contributes to a more
efficient laboratory by centralizing the management of consumable parts.

2.1.2. A New Benchmark of Efficiency


The autosampler’s ultra-fast injection cycle and the ability to input thousands of samples
into the plate-changers (up to 16,896 samples when using 384-well MTPs) allows for
ultra-high-speed multi-sample analysis without requiring the user to replace plates
manually. Standard autosampler functions for automated pretreatment and mobile phase
blending improve the overall efficiency of analysis and method development, and ACTO
functions support simple method transfer.

2.1.3. A New Benchmark of Design


The new Nexera series retains the 260mm width of the previous modular model;
however, the height of the autosampler has been reduced, and the slim oven and newly-
designed dial pump have reduced the LC’s installation space and energy consumption.
Power consumption in standby mode is reduced by up to 81%.
In addition, previously-available features have been improved through new designs.
An auto-cooling function results in zero condensation on the autosampler, and triple
temperature-control in the optical system ensures a more stable baseline than ever. The
PDA shows increased linearity over a wide range and a UV filter improves quantitation
results for photodegradable compounds.

2.3. New Nexera lineup


The new Nexera series has four models:
 Nexera X3 model (maximum pressure 130 MPa)
This is Shimadzu’s new flagship UHPLC model. The dual pump reduces the
gradient delay volume and creates a high pressure gradient, enabling ultra-high-
speed analysis using a packed column with a particle size of under 2 μm. This is
ideal for large-batch analysis and MS coupling. A low pressure gradient can be
achieved by installing an LPGE valve.
 Nexera XS model (maximum pressure 105 MPa)
This is a model designed with competitors’ LC specifications in mind (see Fig. 2
below). It is possible to use a low pressure gradient, the same as that of
competitors, or to install two pumps for a high pressure gradient. There is no dual
pump.
 Nexera XR model (maximum pressure 70 MPa)
This is Shimadzu’s new standard UHPLC model. It can be used both for analysis
with columns with >3μm particle size and for ultra-fast high-level separation
analysis with core-shell columns with less than 3μm particle size. The dual pump
reduces the gradient delay volume and creates a high pressure gradient for ultra-
fast large-batch analysis. A low pressure gradient can be achieved by installing an
LPGE valve. The LC can also be used coupled to an MS.

 Nexera lite model (maximum pressure 44 MPa)
This model is for conventional HPLC analysis with columns or core-shell
columns with a particle size of >3μm. Unlike the other 3 models, the standard
needle for the autosampler is uncoated. If a coated needle is required, it should be
ordered separately. This model will not be actively promoted on the company
website or in catalogues. Materials and information shall be provided to individual
customers at the discretion of the salesperson.

Mixer A gradient mixer with a "mixer device" that
provides automatic volume recognition. The
mixer capacity is automatically recognized by
connecting to the port in the CTO storage with
the optional CMD cable, and it is displayed and
recorded on LabSolution. The 40 μL size
recommended as a low pressure gradient mixer
for XR, XS, and X3 is supplied with a pump
internal mounting plate. ※ However, please
select a larger size when sufficient mixing is
required, such as when using THF.
For high pressure gradients:

Reservoir tray Necessary when using CBM-40/40lite use. Up to


eight 1 L bottles are stored

Mobile phase This device measures the remaining mobile


monitor phase volume in real time. The measurement
data can be checked via the system controller,
LabSolutions, or dedicated software included in
the mobile phase monitor. In coordination with
the Nexera series, mobile phase levels are
monitored in real time and pumping is stopped
when the remaining volume is insufficient. Prior
to starting the batch analysis, the remaining
volume is checked against the volume required
for analysis, and if there is a shortage then a
warning is displayed to alert the user. The
monitoring system consists of a controller and
bottle holders for holding two 1L bottles, which
are set up for use on the reservoir tray. This
optional monitoring system can also be used with
previous LC models or competitor’s products (in
these cases, it is not possible to check whether
mobile phase levels are sufficient prior to batch
analysis).
Controller:
A device to collect sensor signals and send them
to the cloud server. Up to six bottle holders can
be connected.

5. Handling of Existing Products

6. Instrument Installation Guidelines


・Instrument Units (shared by all instruments)
Width: 400 mm (PDA), 530 mm (UV); Depth: 500 mm; Height: 770 mm (max.)
Size The SIL-40C XR and plate changer have a protrusion from the back of 140 mm.
For the plate changer, the value is the width plus 170 mm.
When installing this product, consider the total weight including each instrument, the

Power
Install it at a site where the power supply is easy to control.
Supply
*1: Ensure that the desk or stand on which this product is installed is capable of withstanding the total weight
of the equipment. Make sure that it is flat, stable, and has a depth of at least 700 mm.
*2: Keep the back of the instrument at least 100 mm from the wall. If these conditions are not satisfied, there
is a risk that heat might not be dissipated sufficiently through the slits or by the cooling air from the fan,
resulting in overheating of the instrument and a loss of performance.
*3: Ensure that the indoor space is well ventilated. The solvents used by the high-performance liquid
chromatograph are flammable and toxic. A lack of ventilation may lead to poisoning or a fire.
*4: Install this product near a wash stand or other water supply facilities. If solvent gets in the eyes, or
someone touches a toxic solvent, flush it away immediately with water.
*5: Never use open flames near this product. Do NOT install other units that generate sparks within the same
room. The high-performance liquid chromatograph uses large amounts of organic solvents, and a spark
could cause a fire. To be safe, install a fire extinguisher.
*6: In order to maintain system operating life and performance, do NOT install it at a site exposed to
corrosive gases, organic solvents, halogenated substances, gases containing siloxanes, oil mist, or large
amounts of dirt or dust.
*7: Do NOT install it near equipment that generates strong magnetic fields. In addition, if there is a lot of
noise on the power line, take measures for noise blocking (such as adding a noise filter).
*8: When moving the system, do NOT grip it by the opening/closing part on the front. If the system is
inadvertently lifted up while gripping the opening/closing part on the front, there is a risk that it could be
damaged, or come loose.

Names in the New Nexera Series


The name for the new Nexera series, and the names for the models and units are in
accordance with the following rules.
