Temperature Compensation
The recommended measurement temperature is between 20° and 25° C. However, measurement
can be performed at temperatures between 10°and 30 °C, thanks to the AX-4280’s compensation
function.
Total Diagnostics
With Color-tone & Turbidity Measurement, and Abnormal Color Detection, total urinalysis is determined
automatically. Diagnostics by visual check is no longer necessary.
Color-tone measurement
In addition to the regular test items, total of 23 color-tone is measured for each sample. The
measurement results represent tone and hue.
Turbidity measurement
Turbidity measurement function is added to the parameters. The results will be obtained as Clear, Turbid
and Excessively Turbid.

Normal Measurement
Samples loaded on the Sample Racks are measured in turn. A maximum of 50 or 100 samples
(depending on the rack loading direction) is measured at one time.
STAT Measurement
By interrupting Normal Measurement, emergency test of one sample or several samples can be
performed. To measure only one sample, use STAT Port, and to measure several samples (as
many as 7 samples), use STAT & Control Rack.
After STAT Measurement is completed, Normal Measurement is resumed.
Control Measurement
Measurement accuracy can be maintained by performing Control Measurement. Low, medium, and
high control solutions may be used.
Check Measurement
To check whether the analyzer is working properly, Check Measurement using Check Strips
provided should be performed. There are 2 grays strips and 2 white strips contained in the Check
Strip Case.

4) Principles of measurement
Measurement of test strips is done by the Dual Wavelength Reflectance Method, and specific
gravity is measured by the Index Reflectance Method. The principles of measurement are described
below.
In the Optical Unit, two different ranges of wavelength from the multi-LED are irradiated on the
reacted parts of the test strip and the color-tone-compensated section. These reflected lights are
received by the 3 detectors. At that time, the combination of the wavelengths is different for each
measurement item, as shown in the table below.
Light irradiated to the color-tone-compensation section is adjusted for interfering factors such as
amount of reflected light and color of sample.
From the amount of reflected light measured in the Optical Unit, the reflectance is calculated using
the following formula:
R = Tm ・ Cs / Ts ・ Cm
R: Reflectance
Tm: Reflective light amount with the meas. wavelength
Ts: Reflective light amount with the ref. wavelength
Cm: Reflective light amount at the color-tone-compensated section with the meas.
wavelength
Cs: Reflective light amount at the color-tone-compensated section with the ref. wavelength
However, the calculation of measurement item BLD is made for a single wavelength using the
following formula:
R = Tm / Cm
Reflectance R obtained in the formula is referred to the programmed calibration curve, and is output
as the measurement result.

Finally, in order to compensate for temperature, the reflectance R is corrected using the following
formula:

Specific gravity of urine is obtained by measuring the refraction angles of light that passes through
the Triangle Prism, which contains the urine sample. This is called the Refractive Index Method.
A light from LED (light-emitting diode) becomes a sliver of light through a slit and lenses. The sliver
of light goes through the Triangle Prism Cell that contains the sample, then on to the detector. The
refractive index changes according to the specific gravity of sample in the Triangle Prism Cell. Thus
the angle of the light with respect to the detector changes.
*Position Coefficient: Calculated from the data output by the detector (has a straight-line relationship
with the reflective index).
The reflective index changes according to the temperature of the sample. The value of specific
gravity is corrected using the following formula:
If a large amount of glucose or protein is contained in the urine, the specific gravity is affected.
Therefore, with this analyzer, specific gravity is corrected by using the value of glucose and protein
concentration obtained by measurement with test strips.

Measurement of Color-tone
Total of 23 color-tone is analyzed simultaneously. The chart right shows the combination of tone and
hue.
The tone and hue of urine sample are obtained by different measuring reflectances; R (635 nm), G
(565 nm), B (430 nm), and IR (760 nm).
Y: Reflectance of 430 nm
M: Reflectance of 565 nm
C: Reflectance of 635 nm

4.2.1.4 LED Buzzer Test (Adjust - Hard test - LED & buzzer test)
Choose LED & buzzer test to check the operations concerning the Ready status, measurement,
error trouble LEDs, buzzer, and hand-held barcode reader. This test enables the analyzer to accept
input signals from the hand-held barcode reader. The LEDs will blink, and the alarm sound for one
minute. Press STOP to turn off the LEDs and alarm, and go back to the Hard test menu screen.

To adjust the pulses, press 7 to add 1 pulse, 4 to subtract 1 pulse, 9 to add 10 pulses, and 6 to
subtract 10 pulses. For Wash FB (nozzle washing position, forward and rear), press 5 to raise
the nozzle 10 pulses, and 8 to lower it 10 pulses. Press ENTER to complete your entry and go
to the driving position selection screen. To abort your changes, press ESC to go to the driving
position selection screen. The following table lists the units and the driving positions you can

When On is selected, the check is completed successfully if a sample is detected. No sample will
appear as the measurement result if no sample is detected. When Off is selected, all measurements
will be made normally regardless whether or not a sample exists.
n Test Strip check with the Test Strip Sensor (Strip check)
Specify whether to run the Test Strip check with the Test Strip Sensor at the sampling position. The
available options are On (run) and Off (not run).
When On is selected, the check is completed successfully if a Test Strip is detected. T021 trouble
will appear if no Test Strip is detected. When Off is selected, all measurements will be made
normally regardless whether or not a Test Strip exists.
When On is selected, E040 error will occur if no Washing Solution is detected. When Off is selected,
no error will occur regardless whether or not the Washing Solution exists.
When Off is selected, the analyzer will operate normally and ignores any abnormal specific gravity,
turbidity, or Hb. When On is selected, T063/T064/T065 trouble will be displayed in the event a
related problem arises.

Using the screw driver, adjust VR01 on the Sensor Relay PWA so that the digital voltmeter
reads a value that meets the provisional adjustment standard.
NOTE: If the meter does not read a value meeting the standard shown above when
VR01 is turned fully clockwise, the sensor position may deviate from the Test Strip.
Adjust the pulse to rotate the drum to the position where the meter value meets the
standard. For pulse adjustment, use the following keys:
4: -1 pulse
6: -10 pulses
7: +1 pulse
9: +10 pulses
10) Adjust the pulses by pressing any of 4, 6, 7, and 9, to rotate the drum until the digital voltmeter
reads the maximum value.
NOTE: In the case the digital voltmeter reads 4.5 V or higher as the maximum value,
adjust VR01 on the Sensor Relay PWA using the screw driver so that the value meets
the standard shown in step 9.

11) Keeping the drum at the position determined in step 10, adjust VR01 on the Sensor Relay
PWA using the screw driver, so that the value meets the following adjustment standard:

1 ) After completing 1-1 Test Strip Presence Detection Sensor Sensitivity Adjustment, connect the
- side of the digital voltmeter to the TP05 check terminal on the Sensor Relay PWA, and the +

2 ) Adjust VR02 on the Sensor Relay PWA using the screw driver, so that the digital voltmeter
reads a value that meets the provisional adjustment standard.
3 ) Adjust pulse by pressing any of 4, 6, 7, and 9, to rotate the drum until the digital voltmeter

1 ) Set the data format for Normal Measurement results to Reflectance.
To do that, on the menu on page 4/5 in the Operation mode, set Sample data of Meas
conditions to Reflex.
2 ) Set 10 Sample Tubes containing the Ion Exchange Water in the Rack. Perform Normal
Measurement on these samples.
3 ) Obtain the average C/M and Y/M, and set the Color tone setting.
Go to the Service mode. Display Measure info on page 3/4, and then Color tone on page 2/2.
Use the following formula to obtain the value you should enter.
Displayed C/M value: 1000 (in the case of the default setting)
Value to be entered =1000+(0.003×1000)=1003
Displayed Y/M value: 1002
Value to be entered=1002+(-0.005×1000)=997
NOTE: If the displayed value has been set before, apply that value to the formula.
4 ) Perform Normal Measurement on the ten samples and check if the result falls within the
adjustment standard range given in 5.1.

mm) there. Lightly pressing the divider against the drum, tighten the screw.
NOTE: Divider A has a cutout at the center of its dividing surface. Avoid this cutout
when inserting the gauge.
4 ) For divider 1 adjustment, insert the Clearance Adjustment Jig into the position marked with a
circle. Then, adjust the divider in the same way as above.
5 ) For divider 2 adjustment, insert the Clearance Adjustment Jig. Adjust the divider as you press
on it with tweezers.
n Confirmation
Keeping the drum slot just in front of the divider, check if the 0.07-mm gauge smoothly passes
through the clearance.
Keeping the drum slot just in front of the divider, check that you have a difficulty passing the
0.09-mm gauge through the clearance.
Insert a right-side Test Strip (9EA) in the drum slot and check that the Test Strip passes through
the divider smoothly.
Insert a reversed Test Strip (9EA) in the drum slot and check that the Test Strip passes through
the divider smoothly.
Insert a reversed Test Strip (9EA) in the drum slot and put a right-side Test Strip on it. Check that
only one of these strips can pass through the divider.
NOTE: Use only new Test Strips which have just been taken out of the bottle.
If any of the conditions listed above are not satisfied, it may be because of inadequate divider
adjustment, improper depth of the drum slot, or incorrect width of the shim attached to the drum
slot. Check these points again.

1 ) Lock the Test Strip Loading and Test Strip carrier to the base.
2 ) Lock down the fixing plate (2) (49 mm away from the base edge), and temporarily lock down the
Optical System Unit using two screws (A) as shown below.

3 ) Set the tray and place a Test Strip in the slot just in front of the optical metering position (as
appropriate).
4 ) Loosen all the screws that lock down the optical system. Position the unit to satisfy the
conditions shown below and temporarily lock it down using the screws (A). During adjustment,
the Chuck Assembly must be placed to the right or left.
(1) As seen from above the Optical System Unit, the Test Strip should be parallel to
the Optical System Shaft.
(2) The center of the Test Strip Jig should match that of the chuck.
5 ) Connect the carrier motor to a 12 V power supply or the analyzer (in the case of the analyzer,
PI must also be connected), and use the chuck to carry the Test Strip to one stage before the
optical metering position.
6 ) Check the Test Strip and Optical System Shaft are parallel, again. If not, adjust the unit position
again. After positioning is completed, tighten all the screws on the Optical Unit.
NOTE: This confirmation must be made with all the screws on the Optical System Unit
tight. The Carrier Cover must also be locked.
Preparation:
Ground anywhere between the Test Strip Carrier Base and the analyzer frame. Attach the carrier
and Optical System to the analyzer for Check Measurement.
1 ) In the Service mode, press MENU twice. Choose 2: System option, and then 1: Measure.
2 ) While the fifth item Check result appears, press - several times to select Detail, and press
ENTER.
3 ) Press MENU twice. Choose 2: Adjust, 2: Pulse & LED, and then 1: LED Adjust.
4 ) While LED Adjust appears, press - several times to select OPT, and press ENTER.
7 ) When the Check Strip comes to the Optical Block, press START.
8 ) After Optical Block movement, rotate VR6 on the Main PWA using the screw driver to adjust the
light amount for Test Strip pad detection.
Confirmation:
1 ) Turn on the power to the analyzer. Make sure the Ready screen appears, and then press
CHECK.
2 ) Place a Check Strip (white) on the tray and press START to make a Check Measurement.

rib at the back of the tray.)
5 ) Retry the operation from step 1. Before the Check Strip is carried to the Optical Block, remove
the Check Strips that have passed the Test Strip Pass Sensor from the tray (so that the Test
Strip set in step 4 is measured).
6 ) Check the measurement result satisfies the confirmation standard shown in step 3.
7 ) Do the same operation as in step 4 on the opposite side (the left-front side of the strip contacts
the left edge of the slot on the rib in front of the tray, and the right-back side contacts the right
edge of the slot on the rib at the back of the tray). Check the result satisfies the standard
shown in step 3.
If any of the three measurement results does not meet the standard, retry adjustments.

(4) Choose 2: Unit action.
(5) Choose Rack.
3 ) To make adjustments:
(1) Choose Init to initialize the system.

position.

meeting the standard. If you replace the shim, run the motor again to bring the seam of the belt
to the screw that locks down the top base.
7 ) Write the resulting thickness of the shim on a label, and attach it to the shim.
