    Sertoli cells (SC) have been used for their immunomodulatory properties in cell transplants (Emerich et al., 2003). Moreover, SC themselves may prevent immune rejection (Sipone et al., 2006) and possess a natural phagocytic activity: the latter may make them suitable as biocompatible drug delivery carriers. Porcine SC were loaded with PLA microspheres containing pharmaceutical agents (SC-MS). Phagocytosis was monitored over 24 hours; the uptake was measured by HPLC at fixed time points and followed up through 6 days. SC viability and morphology were monitored together with reactive oxygen species (ROS), DNA damage and parameters of SC functionality and immunomodulatory properties over time. A preliminary antibacterial activity was assessed in vitro. SC-MS were cryopreserved in liquid nitrogen and after plating underwent the same characterization.     The comet assay or single cell gel electrophoresis, is the most common method used to measure strand breaks and a variety of other DNA lesions in human populations. To estimate the risk of overall mortality, mortality by cause, and cancer incidence associated to DNA damage, a cohort of 2,403 healthy individuals (25,978 person-years) screened in 16 laboratories using the comet assay between 1996 and 2016 was followed-up. Kaplan–Meier analysis indicated a worse overall survival in the medium and high tertile of DNA damage (p < 0.001). The effect of DNA damage on survival was modelled according to Cox proportional hazard regression model. The adjusted hazard ratio (HR) was 1.42 (1.06–1.90) for overall mortality, and 1.94 (1.04–3.59) for diseases of the circulatory system in subjects with the highest tertile of DNA damage. The findings of this study provide epidemiological evidence encouraging the implementation of the comet assay in preventive strategies for non-communicable diseases.

    A travel medicine clinic is the proper medical centre aimed to provide updated recommendations before any journey. The study aim was to assess the number of accesses during 2016 at the Travel Medicine Clinic of the Local Health Unit in the Umbria Region. An electronic ad hoc database was developed. Paper-based data referring to 2016 were recorded in this electronic registry, developed on Microsoft Office Access®. In 2016, 891 subjects came in our clinic in order to get information before travelling. In our sample, 53.3% were male and the mean age was 35.33 years ± 0.58. Almost half had a higher education, and 20% were foreign. Vaccines against food- and water-borne diseases were the most frequently administered.     Recent data support the hypothesis that genetic damage occurring early in life during childhood can play an important role in the development of chronic diseases in adulthood, including cancer. The objective of this paper, part of the MAPEC_LIFE project, is to describe the frequency of micronuclei and meta-nuclear alterations in exfoliated buccal cells of 6-8year-old Italian children recruited in five Italian towns (i.e., Brescia, Torino, Pisa, Perugia and Lecce) with different air pollution levels. About 200 children per town were recruited from primary schools. Biological samples were collected twice from the same children, in two different seasons (winter 2014-15 and late spring 2015).     The aim of this work was to characterise Lenticchia di Castelluccio di Norcia (an Italian PGI lentil landrace) and assess the impact of cooking and storage on antioxidant activity. After opening the package (T0), samples were analysed using a set of chemical assays (i.e., total phenolic content, DPPH, ABTS, and ORAC assays). Analyses were also conducted on boiled, pressure-cooked, and 6-month-stored (T1) products. At both T0 and T1, raw Lenticchia di Castelluccio di Norcia PGI showed higher total phenolic content (T0: 9.08 mg GAE/g, T1: 7.76 mg GAE/g) and antioxidant activity (DPPH T0: 33.02 µmol TE/g, T1: 29.23 µmol TE/g; ABTS T0: 32.12 µmol CE/g, T1: 31.77 µmol CE/g; ORAC T0: 3.58 μmol TE/g, T1: 3.60 μmol TE/g) than boiled and pressure-cooked samples.     Phenylselenenylzinc chloride (PhSeZnCl) is an air-stable selenolate, easily synthesizable through oxidative insertion of elemental zinc into the Se-halogen bond of the commercially available phenylselenyl chloride. PhSeZnCl was shown to possess a marked GPx-like activity both in NMR and in vitro tests, and to effectively react with cellular thiols, and was supposed for a potential use in the chemotherapy of drug-resistant cancers. However, activity of PhSeZnCl in hepatic cells has never been tested before now. In this in vitro approach, we evaluated the cytotoxic, genotoxic, and apoptotic activities, as well as the effects on cell cycle of PhSeZnCl in two preclinical hepatic models, namely HepG2 and HepaRG cells.     The aim of this work was to evaluate in vitro the genotoxic and/or antigenotoxic effects of resveratrol (RESV) and pterostilbene (PTER) on HepG2 cells. Moreover, additional tests were performed to evaluate early and late apoptosis events induced by the tested stilbenes. RESV and PTER did not show any genotoxic activity. As regards antigenotoxicity testing, RESV and PTER showed a typical, U-shaped hormetic dose-response relationship characterized by a biphasic trend with small quantities having opposite effects to large ones. HepG2 cells treated with PTER exhibited a marked increase in early apoptosis (40.1 %) at 250 μM; whereas, the highest concentration tested for both RESV and PTER significantly increased the proportion of HepG2 cells undergoing late apoptosis (32.5 and 51.2 %, respectively).     Breast cancer (BC) is the most common cancer in women and several factors are involved in its onset. Green tea (GT) has been shown to have potential beneficial effects on different types of cancer. The aim of this review was to evaluate the association between GT regular consumption and risk of BC in women. The risk of BC recurrence and risk of BC in relation to menopausal status were also evaluated. A literature search of PubMed, Scopus, and Web of Science was conducted. Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines were followed to perform the systematic review and meta-analysis. Full texts were downloaded for 40 studies; however, only 13 records were included in the meta-analysis. Eight were cohort studies and five were case-control studies.     Air pollution in urban areas is a major concern as it negatively affects the health of a large number of people. The purpose of this study was to assess the inhalation health risk for exposure to PM and benzene of the populations living in three Italian cities. Data regarding PM and benzene daily measured by "traffic" stations and "background" stations in Torino, Perugia, and Lecce during 2014 and 2015 were compared to the limits indicated in the Directive 2008/50/EC. In addition, an inhalation risk analysis for exposure to benzene was performed for adults and children by applying the standard United States Environmental Protection Agency's (USEPA) methodology. The levels of PM detected in Torino exceeded the legal limits in both years with an increased mean concentration >10 µg/m³ comparing with background station. Included studies were of the case-control studies type. From an initial list of 643 potentially eligible articles, seven studies were finally retained in the quantitative analysis. This work was performed according to the Cochrane Collaboration and the PRISMA guidelines and recommendations. Articles published before November 2021 were identified by an advanced search on PubMed/MEDLINE, Scopus and Web of Science databases. A critical appraisal using a specific tool was conducted to assess the quality of each included study. All analyses were carried out by using the Review Manager (RevMan) 5.4 software (The Cochrane Collaboration, London, UK).     The comet assay is widely used in human biomonitoring to measure DNA damage as a marker of exposure to genotoxic agents or to investigate genoprotective effects. Studies often involve small numbers of subjects, and design may be sub-optimal in other respects. In addition, comet assay protocols in use in different laboratories vary significantly. In spite of these difficulties, it is appropriate to carry out a pooled analysis of all available comet assay biomonitoring data, in order to establish baseline parameters of DNA damage, and to investigate associations between comet assay measurements and factors such as sex, age, smoking status, nutrition, lifestyle, etc. With this as its major objective, the ComNet project has recruited almost 100 research groups willing to share datasets. Here we provide a background to this project, discussing the history of the comet assay and practical issues that can critically affect its performance. We survey its diverse applications in biomonitoring studies, including environmental and occupational exposure to genotoxic agents, genoprotection by dietary and other factors, DNA damage associated with various diseases, and intrinsic factors that affect DNA damage levels in humans. We examine in depth the quality of data from a random selection of studies, from an epidemiological and statistical point of view.

    Air pollutants, especially PM, have been found to determine various effects on human health, including genotoxic effects. The aim of this study was to assess DNA damage with micronuclei (MN) and comet tests on buccal cells of 6–8 years old children living in an area with high air pollution. Both tests were repeated in the same children in two consecutive winters to compare the levels of DNA damage under different pollution conditions. A complete data set including lifestyle, air pollutants levels and biological sampling was available for 180 children in the two winters. A high mean MN frequency was found in both seasons, with higher value in the first (0.51 ± 0.59) than the second winter (0.40 ± 0.52), whereas DNA damage measured with comet test showed higher damage in the second versus the first winter (visual score 208.8 ± 67.1 vs. 173.2 ± 50.8).     Road tunnel construction involves the use of explosives and diesel-powered machines in a very limited indoor space. Tunnel construction workers are thus exposed to a variety of toxic substances, including dust, asbestos, silica, concrete, diesel fumes, and oil mist. In this study, the low noise miniaturized Sioutas Cascade Impactor (SKC) has been used to collect PM samples as a function of particle size (i.e., aerodynamic diameter). Airborne particulates were sampled into a road tunnel (final length 2391 m) under construction near the village of Pale, Municipality of Foligno, Umbrian Apennines (State Highway 77 “Val di Chienti”). Three sampling sessions were performed: (i) in the West yard, during dislodging of rocks; (ii) in the East yard during drilling of rocks, and (iii) during scaling and grouting with quick-drying shotcrete. Toxicological evaluation was carried out in vitro on A549 lung carcinoma cells: cytotoxicity was assessed by Trypan blue dye exclusion assay, whereas genotoxicity was assessed by comet assay (primary DNA damage) and cytokinesis-block micronucleus (CBMN) Cytome assay (cytogenetic effects). Primary DNA damage (i.e., strand breakage) was mainly caused by coarse fractions A (Ø > 2.5 μm) sampled in the West yard (sample i) and in the East yard (sample iii). Cytogenetic effects were mainly caused by the fine fractions AF (aerodynamic Ø < 0.25 μm) sampled in the East yard (sample ii). Results reported in this article show that particulate matter (PM) samples collected underground (i.e., during road tunnel construction) may trigger various cytotoxic/genotoxic effects. The recent classification of air pollution and PM itself as carcinogenic to humans (group 1) by the IARC makes indoor/outdoor air pollution a growing matter of concern. Our results highlight the need in this occupational setting for a reduction, if applicable, of PM emission (prevention), or an implementation of work practices by stressing the importance of proper use of protective equipment (protection).

    Many studies have shown the presence of numerous organic genotoxins and carcinogens in drinking water. These toxic substances derive not only from pollution, but also from the disinfection treatments, particularly when water is obtained from surface sources and then chlorinated. Most of the chlorinated compounds in drinking water are nonvolatile and are difficult to characterize. Thus, it has been proposed to study such complex mixtures using short-term genotoxicity tests predictive of carcinogenic activity. Mutagenicity of water before and after disinfection has mainly been studied by the Salmonella/microsome (Ames test); in vitro genotoxicity tests have also been performed in yeasts and mammalian cells; in situ monitoring of genotoxins has also been performed using complete organisms such as aquatic animals or plants (in vivo).
