The inducible transcription factor nuclear factor-kappaB (NF-kappaB) plays an important role in the regulation of immune, inflammatory and carcinogenic responses. While normal NF-kappaB activation is necessary for cell survival and immunity, deregulated NF-kappaB expression is a characteristic phenomenon in cancer development, as well as in several inflammatory diseases. Hence, NF-kappaB has become a major target in drug discovery, and several natural and synthetic compounds have been investigated for their potential to inhibit NF-kappaB. Here, we discuss the applications of marine natural products, in particular, as novel, potent NF-kappaB inhibitors. With the oceans covering two-thirds of the Earth's surface, and with the uniqueness of the environmental properties of marine habitats, it is easily understandable that organisms thriving in the oceans constitute a rich source of chemically unique and biomedically powerful secondary metabolites. Since the early 1960s, significant effort has been placed on the pharmacological evaluation of marine secondary metabolites. Noteworthy achievements of this field of biomedically guided marine exploration, a scientific endeavour often referred to as the search for "Drugs from the Sea", include the discovery of numerous potent anti-cancer, anti-inflammatory, antimicrobial, and analgesic compounds. The chemical characteristics and molecular targets of marine NF-kappaB inhibitors discovered to date are presented and discussed in the context of marine chemical ecology.

Development of chemoresistance in androgen-refractory prostate cancer cells is partly due to constitutive activation of Rel/NF-κB transcription factors that regulate several cell survival and anti-apoptotic genes. In this study we examined whether betulinic acid (BetA), a pentacyclic triterpene from the bark of white birch, is effective in inhibiting NF-κB expression in androgen-refractory human prostate cancer cells exhibiting high constitutive NF-κB expression. Treatment of PC-3 cells with BetA inhibited DNA binding and reduced nuclear levels of the NF-κB/p65. BetA-mediated NF-κB inhibition involved decreased IKK activity and phosphorylation of IκBα at serine 32/36 followed by its degradation. Reporter assays revealed that NF-κB inhibition by BetA is transcriptionally active. These effects were found to correlate with a shift in Bax/Bcl-2 ratio and cleavage of poly(ADP)ribose polymerase more towards apoptosis. BetA also inhibited TNFα-induced activation of NF-κB via the IκBα pathway, thereby sensitizing the cells to TNFα-induced apoptosis. Our studies demonstrate that BetA effectively inhibits constitutive NF-κB activation and supports the rationale for targeting NF-κB through combination protocols with BetA in androgen-refractory prostate cancer. © 2008 Wiley-Liss, Inc.

Hepatic myofibroblast activation during injury causes deposition of extracellular matrix within the liver and promotes development of fibrosis. Hepatic myofibroblast apoptosis is associated with remodelling of fibrotic extracellular matrix and regression of fibrosis. Previous work showed that inhibition of constitutive NF-κB signaling promotes hepatic myofibroblast apoptosis and resolution of fibrosis in rodent models. However, to date agents used to target constitutive NF-κB transcriptional activity in hepatic myofibroblasts have been relatively non-specific with potential for off-target effects that may complicate data interpretation. Likewise, rat chronic liver disease models may not accurately recapitulate the activation of human hepatic myofibroblasts.We used a mutant recombinant IκBα super-repressor fused to the HIV–TAT domain to specifically target NF-κB signaling in hepatic myofibroblasts. Inhibition of NF-κB activity was measured using reporter assay. Apoptosis of hepatic myofibroblasts was assessed by morphological changes, cleavage of the PARP-1 protein and Caspase 3 activation.TAT-IκBαSR reduced NF-κB dependent transcription, Bcl-2 expression and promoted Jun-N-terminal kinase-dependent apoptosis in human and rat hepatic myofibroblasts.These data highlight the conserved role of NF-κB during fibrogenesis. Our data validate the use of rodent models for pre-clinical testing of NF-κB inhibitors as anti-fibrotics and stimulators of fibrotic extracellular matrix remodelling.

NGF treatment of neuroblastoma cells stimulates outgrowth of neurite processes associated with the expression of TrkA receptor and several differentiation markers. In this study, a 6 DIV exposure to NGF (50 ng/ml) increased immunostaining for alpha-tubulin, and expression of both alpha-tubulin and protein kinase C in the neuroblastoma cell line Neuro2a. Further, up-regulation of transglutaminase 1 and transglutaminase 2 expression, and reduction of transglutaminase 3 levels, were also observed in NGF-treated cells in comparison to untreated cells. Moreover, when Neuro2a cells were treated with the specific NF-kappaB inhibitor SN-50, the strong reduction of NF-kappaB activation was concomitant with a significant decrease of transglutaminase 2 expression, suggesting that NGF-evoked transglutaminase 2 induction could be related to NF-kappaB activation. To characterize the possible transglutaminase 2/NF-kappaB interplay, NGF treatment was carried out in Neuro2a cells which already over-expressed transglutaminase 2 after retinoic acid treatment. An additive effect of NGF was observed on the retinoic acid-induced transglutaminase 2 expression and enzyme activity, and NF-kappaB activation. However, a cystamine-mediated significant inhibition of transglutaminase activity (70%) was accompanied by a drastically reduced NF-kappaB activation only in cells exposed to NGF following retinoic acid treatment. We hypothesize that NF-kappaB activation was dependent on the transamidating activity related to high levels of TG2, and NGF enhanced NF-kappaB activation by a different, synergistically acting, pathway. These data suggest that the combined use of NGF and retinoic acid, or mimicking drugs, may provide the basics for the development of novel strategies in the therapeutic management of neuroblastomas.
