Pyrrolizidine Alkaloids

Following scientific risk assessments, several countries have imposed strict regulations on herbal medicines containing 1,2-dehydro-pyrrolizidine alkaloids. Using published data on the plants used in honey production, pyrrolizidine alkaloid-containing plants are shown in this review to represent a significant source of honey worldwide. This observation, honey consumption data, reported levels of pyrrolizidine alkaloids in honeys, and consideration of tolerable exposure levels determined for pyrrolizidine alkaloids in herbal medicines, leads to the conclusion that some honey is a potential threat to health, especially for infants and fetuses, and further investigation is warranted.

Comfrey (Symphytum L.) is used to treat bone fractures, tendon injuries, ulcer lesions of gastrointestinal tract. It promotes wound healing, accelerates exudates resorption in lungs and reduces joints' inflammation. In Georgian folk medicine, herbal remedies from comfrey are used to accelerate regeneration processes. Comfrey contains hepatotoxic and carcinogenic pyrrolizidine alkaloids, besides the main active ingredient is poly [3 - (3,4-dihydroxyphenyl) glyceric acid] (PDPGA). The aim of present work was to develop a technology for the substance - poly [3-(3,4dihydroxyphenyl) glyceric acid] (PDPGA) from comfrey stems, free of toxic pyrrolizidine alkaloids. This article aimed 1) to review herbal medicine containing pyrrolizidine alkaloids (PA)-induced toxicities of the liver; 2) to encourage the recognition and prevention of common problems encountered when using complementary and alternative medicine and 3) to review the toxic effects of herbal remedies containing PAs. We performed a systematic literature search using the PubMed and Google Scholar engines. The search was not restricted to languages. We also provide an interpretation of the data. Herbal remedies containing PAs can induce liver damage, including hepato- sinusoidal obstruction syndrome or veno-occlusive disease. Preventing overdose and monitoring long-term use of such remedies may avoid glutathione depletion leading to mitochondrial injury, and therefore avoid liver cell damage. Alkaloid profiles of five Senecio species (Asteraceae), including S. ambiguus subsp. ambiguus, S. ambiguus subsp. nebrodensis, S. gibbosus subsp. bicolor, S. gibbosus subsp. gibbosus, and S. gibbosus subsp. cineraria, were studied. Eleven pyrrolizidine alkaloids were identified and their content was evaluated by GLC-MS and GLC analysis. Otosenine and florosenine were found to be the major alkaloids in all studied species. It is interesting that only S. ambiguus subsp. nebrodensis was characterized by a high content of the alkaloids jacobine, jacoline, jaconine, and jacozine.

A novel pyrrolizidine alkaloids, madhumidine A (1), and two known alkaloids, lindelofidine benzoic acid ester (2) and minalobine B (3) were isolated from the leaves of Madhuca pasquieri (Dubard) H. J. LAM. The chemical structures of these alkaloids were established mainly by NMR techniques and mass spectrometry. Their anti-inflammatory activity was evaluated against lipopolysaccharide-induced nitric oxide production in macrophage RAW264.7 cell. In addition, the cytotoxic activity of all isolated compounds was tested against a panel of cancer cell lines.

The ability of ovine ruminal fluid to metabolize pyrrolizidine alkaloid (PA) from Senecio jacobaea under anaerobic conditions was evaluated. Four fistulated sheep fed PA served as individual sources of ruminal fluid, which was incubated in a defined minimal salts medium under two different anaerobic conditions, denitrifying and methanogenic. Anaerobic cultures amended with ovine ruminal fluids (20%), PA (100 micrograms/ml), and a defined minimal salts medium were monitored for a period of several days. These cultures revealed that while PA was not depleted in sterile, autoclaved controls or under denitrifying conditions, it was metabolized during periods of active methanogenesis under methanogenic conditions. The toxicity assessment of herbal medicines is important for human health and appropriate utilization of these medicines. However, challenges have to be overcome because of the complexity of coexisting multiple components in herbal medicines and the highly interconnected organismal system. In this study, a target profiling approach was established by combining the characteristic fingerprint analysis of herbal chemicals with potential toxicity through a precursor ion scan-based mass spectroscopy and the target profiling analysis of biomarkers responsible for the toxicity. Through this newly developed approach, the comparative hepatotoxicity assessment of two herbal medicines from the same genus, Senecio vulgaris L. and Senecio scandens Buch.-Ham, was performed. Secondary metabolites such as pyrrolizidine alkaloids (PAs) play a crucial part in plant defense. PAs can occur in plants in two forms: tertiary amine (free base) and N-oxide. PA extraction and detection are of great importance for the understanding of the role of PAs as plant defense compounds, as the tertiary PA form is known for its stronger influence on several generalist insects, whereas the N-oxide form is claimed to be less deterrent. We measured PA N-oxides and their reduced tertiary amines by liquid chromatography-tandem mass spectrometry (LC-MS/MS). We show that the occurrence of tertiary PAs is not an artifact of the extraction and detection method. We found up to 50% of tertiary PAs in shoots of Jacobine - chemotype plants of Jacobaea vulgaris. Jacobine and its derivatives (jacoline, jaconine, jacozine and dehydrojaconine) may occur for more than 20% in reduced form in the shoots and more than 10% in the roots. For 22 PAs detected in F(2) hybrids (J. vulgaris × Jacobaea aquatica), we calculate the tertiary amine percentage (TA%=the tertiary amine concentration/(tertiary amine concentration+the corresponding N-oxide concentration) × 100). We found that the TA% for various PAs was genotype-dependent. Furthermore, TA% for the different PAs were correlated and the highest correlations occurred between PAs which share high structural similarity.

Aim/Background: Boraginaceae is famous for the production of PAs and some of these PAs are carcinogenic, and also cause liver failure. So the aim of present study was to identify the presence or absence of hepatotoxic pyrrolizidine alkaloids in the tribe Lithospermea (Boraginaceae). If any are found, it may indicate excluding members of this tribe from herbal formulation or from use on patients with liver problems.
Materials and Methods: Plant samples of Onosma hispida Wall. ex G. Don, Onosma paniculatum Bureau & Franch., Onosma hookeri var. longiflorum (Duthie) A.V. Duthie ex Stapf, and Maharanga emodi (Wall.) A. DC. from Boraginaceae—Tribe Lithospermeae were collected from various regions of Pan Himalaya and brought to Beijing Normal University for further experimentation. We used acetonitrile—water gradient with 0.1% formic acid as the mobile phase and Zorbax SB-Aq column, to analyze samples. Furthermore, we also searched the literature to find ethnomedicinal importance of these plants.
Results: The results showed that these plants are used orally for the treatment of various human ailments and so we further investigated these plants for toxic pyrrolizidine alkaloids (PAs). HPLC results showed that leaves of these plants were PA positive, and out of four PA standards, three: Heliotrine (2), Lycopsamine (3), and Echimidine (4) were detected.
Conclusions: In this study, we present a new report about the presence of toxic PAs in the leaves of Onosma hispida, O. paniculatum, Onosma hookeri var. longiflorum, and Maharanga emodi from the Pan-Himalaya region. These plants are used in traditional medicine mostly in Pakistan, Nepal, and China, and the presence of hepatotoxic PAs limit the use for medicinal purposes

Senecio ambiguus subsp.ambiguus (Biv.) DC. extracts were able to inhibit thein vitro proliferation of renal cell adenocarcinoma ACHN and hormone dependent prostate carcinoma LNCaP. The potential cytotoxic property of the plant was revealed by the methanolic extract action against LNCaP (IC50 of 5.51 μng/mL) and ACHN (IC50 of 38.95 μg/mL). The most potent cytotoxic activity (IC50 of 5.34 μg/mL against the prostate carcinoma cell line) was exerted by the dichloromethane extract. Through bioassay-guided fractionation of the dichloromethane extract jacaranone was isolated as the major active constituent. This quinoid showed a very strong activity against ACHN and LNcaP with IC50 of 4.32 and 7.39 (μg/mL, respectively. Its structure was established by GC/MS and NMR analysis. Then- bexane extract showed an interesting inhibition on the proliferation of tumor cell lines an IC50 value of 5.23 μg/mL against LNCaP. Three compounds identified in then-hexane extract such as nerolidol,a-humulene andg-tocopherol were found to be active aginst LNCAP with IC50 values ranged from 11.24 to 15.56 μg/mL.
