Hidradenitis suppurativa (HS) is a chronic skin disease of the pilo-sebaceous apocrine unit characterized by significant inflammation and an impaired quality of life. The pathogenesis of HS remains unclear. To determine the HS skin and blood transcriptomes and HS blood proteome, patient data from previously published studies were analysed and integrated from a cohort of patients with moderate to severe HS (n = 17) compared to healthy volunteers (n = 10). The analysis utilized empirical Bayes methods to determine differentially expressed genes (DEGs) (fold change (FCH) >2.0 and false discovery rate (FDR) <0.05), and differentially expressed proteins (DEPs) (FCH>1.5, FDR<0.05). In the HS skin transcriptome (lesional skin compared to non-lesional skin), there was an abundance of immunoglobulins, antimicrobial peptides, and an interferon signature. Gene-sets related to Notch signalling and Interferon pathways were differentially activated in lesional compared to non-lesional...

Genetic variants near and within the fatty acid desaturase (FADS) cluster are associated with polyunsaturated fatty acid (PUFA) biosynthesis, levels of several disease biomarkers and risk of human disease. However, determining the functional mechanisms by which these genetic variants impact PUFA levels remains a challenge. Utilizing an Illumina 450K array, we previously reported strong allele-specific methylation (ASM) associations (p = 2.69×10-29) between a single nucleotide polymorphism (SNP) rs174537 and DNA methylation of CpG sites located in the putative enhancer region between FADS1 and FADS2, in human liver tissue. However, this array only featured 20 CpG sites within this 12kb region. To better understand the methylation landscape within this region, we conducted bisulfite sequencing of the region between FADS1 and FADS2. Liver tissues from 50 male subjects (27 European Americans, 23 African Americans) were obtained from the Pathobiological Determinants of Atherosclerosis in...

Antiviral defenses are inappropriately activated in systemic lupus erythematosus (SLE) and association between SLE and the antiviral helicase gene, IFIH1, is well established. We sought to extend the previously reported association of pathogenic soluble mediators and autoantibodies with mouse Mda5 to its human ortholog, IFIH1. To better understand the role this gene plays in human lupus, we assessed association of IFIH1 variants with soluble mediators and autoantibodies in 357 European-American SLE patients, first-degree relatives, and unrelated, unaffected healthy controls. Association between each of 135 genotyped SNPs in IFIH1 and four lupus-associated plasma mediators, IL-6, TNF-α, IFN-β, and IP-10, were investigated via linear regression. No significant associations were found to SNPs orthologous to those identified in exon 13 of the mouse. However, outside of this region there were significant associations between IL-6 and rs76162067 (p = 0.008), as well as IP-10 and rs7971102...

Most investigations to date aiming to identify the genetic basis of the stress response of wheat (Triticum aestivum L.) have focused on the response to single stress agents such as high temperature or drought, even though in the natural situation, these stresses tend often to occur together. Here, a panel of 108 spring type bread wheat cultivars was phenotyped for 15 yield and yield related traits for two years (2014/15 and 2015/16) under non-stressed conditions, under high temperature stress, under drought and under a combined high temperature and drought regime. The mean loss in grain yield caused by all stress regimes was 51.33%. Analysis of variance (ANOVA) of yield trait showed significant differences among genotypes environments and their interactions (G×E). All the studied traits had higher heritability values which ranged from 0.35-0.94 under [C], 0.58-0.95 under [D], 0.62-0.93 under [H] and 0.60-0.95 under [HD]. GWAS was performed by using 9,646 informative SNP markers and ...

Physical activity is known to be preventive against various non-communicable diseases. We investigated the relationship between daily physical activity level and plasma metabolites using a targeted metabolomics approach in a population-based study. A total of 1,193 participants (male, aged 35 to 74 years) with fasting blood samples were selected from the baseline survey of a cohort study. Information on daily total physical activity, classified into four levels by quartile of metabolic equivalent scores, and sedentary behavior, defined as hours of sitting per day, was collected through a self-administered questionnaire. Plasma metabolite concentrations were quantified by capillary electrophoresis mass spectrometry method. We performed linear regression analysis models with multivariable adjustment and corrected p-values for multiple testing in the original population (n = 808). The robustness of the results was confirmed by replication analysis in a separate population (n = 385) cre...

To develop novel interventions for autism spectrum disorder (ASD) core symptoms, valid, reliable, and sensitive longitudinal outcome measures are required for detecting symptom change over time. Here, we tested whether a computerized analysis of quantitative facial expression measures could act as a marker for core ASD social symptoms. Facial expression intensity values during a semi-structured socially interactive situation extracted from the Autism Diagnostic Observation Schedule (ADOS) were quantified by dedicated software in 18 high-functioning adult males with ASD. Controls were 17 age-, gender-, parental socioeconomic background-, and intellectual level-matched typically developing (TD) individuals. Statistical analyses determined whether values representing the strength and variability of each facial expression element differed significantly between the ASD and TD groups and whether they correlated with ADOS reciprocal social interaction scores. Compared with the TD controls,...

The lack of precise biomarkers that identify patients at risk for myocardial injury and stable angina delays administration of optimal therapy. Hence, the search for noninvasive biomarkers that could accurately stratify patients with impending heart attack, from patients with stable coronary artery disease (CAD), is urgently needed in the clinic. Herein, we performed comparative quantitative proteomics on whole plasma sampled from patients with stable angina (NMI), acute myocardial infarction (MI), and healthy control subjects (Ctrl). We detected a total of 371 proteins with high confidence (FDR < 1%, p < 0.05) including 53 preliminary biomarkers that displayed ≥2-fold modulated expression in patients with CAD (27 associated with atherosclerotic stable angina, 26 with myocardial injury). In the verification phase, we used label-free LC-MRM-MS-based targeted method to verify the preliminary biomarkers in pooled plasma, excluded peptides that were poorly distinguished from backg...

Liver and mammary gland are among the most important organs during lactation in dairy cows. With the purpose of understanding both the different and the complementary roles and the crosstalk of those two organs during lactation, a transcriptome analysis was performed on liver and mammary tissues of 10 primiparous dairy cows in mid-lactation. The analysis was performed using a 4×44K Bovine Agilent microarray chip. The transcriptome difference between the two tissues was analyzed using SAS JMP Genomics using ANOVA with a false discovery rate correction (FDR). The analysis uncovered >9,000 genes differentially expressed (DEG) between the two tissues with a FDR<0.001. The functional analysis of the DEG uncovered a larger metabolic (especially related to lipid) and inflammatory response capacity in liver compared with mammary tissue while the mammary tissue had a larger protein synthesis and secretion, proliferation/differentiation, signaling, and innate immune system capacity comp...

Most existing statistical methods developed for calling single nucleotide polymorphisms (SNPs) using next-generation sequencing (NGS) data are based on Bayesian frameworks, and there does not exist any SNP caller that produces p-values for calling SNPs in a frequentist framework. To fill in this gap, we develop a new method MAFsnp, a Multiple-sample based Accurate and Flexible algorithm for calling SNPs with NGS data. MAFsnp is based on an estimated likelihood ratio test (eLRT) statistic. In practical situation, the involved parameter is very close to the boundary of the parametric space, so the standard large sample property is not suitable to evaluate the finite-sample distribution of the eLRT statistic. Observing that the distribution of the test statistic is a mixture of zero and a continuous part, we propose to model the test statistic with a novel two-parameter mixture distribution. Once the parameters in the mixture distribution are estimated, p-values can be easily calculate...

Determining and observing the minimum allowable distance of the marine earth station in motion (M-ESIM) from the shore prevents its destructive interference on the co-frequency shore fixed station. These earth stations are providing broadband Fixed Satellite Services (FSS). This paper studies the parameters involved in the minimum allowable distance of the ship from the shore by simulating the interference from an M-ESIM on a shore fixed station. This distance is determined versus the latitude, carrier frequency and the annual number of crossings ships along the shore. The results show that with decreasing the carrier frequency, decreasing latitude or increasing the number of annual passing vessels, this minimum distance increases. A solution to eliminate these effects and keep constant the minimum distance is shifting the transmitter frequency band. This frequency shift increases the frequency dependent rejection (FDR). In this paper, values of FDR are obtained separately versus th...

Constitutive activation of interferon signaling pathways has been reported in colorectal cancer (CRC), leading to a strong CD8+ T cell response through stimulation of NLRC5 expression. Primed CD8+ T cell expansion, however, may be negatively regulated by PD-L1 expression. Additionally, aberrant PD-L1 expression enables cancer cells to escape the immune attack. Our study aimed to select potential regulatory variants in the NLRC5 and PD-L1 genes by using several online in silico tools, such as UCSC browser, HaploReg, Regulome DB, Gtex Portal, microRNA and transcription factor binding site prediction tools and to investigate their influence on CRC risk in a Czech cohort of 1424 CRC patients and 1114 healthy controls. Logistic regression analysis adjusted for age and gender reported a moderate association between rectal cancer risk and two NLRC5 SNPs, rs1684575 T>G (OR: 1.60, 95% CI: 1.13-2.27, recessive model) and rs3751710 (OR: 0.70, 95% CI: 0.51-0.96, dominant model). Given that a...

The indicator that best expresses the quality of life (welfare) is gross domestic product per capita. The two factors that account for economic growth (number of employees and labor productivity) are taken into account, but the macroeconomic outcome must provide an average living standard for the entire population. The system of national accounts used by the Member States of the European Union as well as by other states as a form of evidence and macroeconomic analysis contains all the elements needed for such a study. In the period under consideration, regardless of the method of calculating GDP, we obtain a precise quantitative amount that, compared to previous developments, suggests how the quality of life of the population has evolved. In analyzing the quality of life, we have to go from the structure of the population, number of employees, other categories, expressed by statistical indicators.

Banks as financial institutions need to maintain their performance in order to operate optimally. Moreover, Islamic banks must compete with conventional banks which are dominant and growing rapidly in Indonesia. This increasingly sharp and tight competition must be accompanied by good and orderly management in order to survive in the banking industry for a long time. One of the factors that must be considered by a bank in order to survive is the bank's financial performance. Comparison of the financial performance of conventional banks and Islamic banks is carried out using financial ratios in the form of the dependent variable ROA and the independent variables CAR, NPL/NPF, LDR/FDR taken from annual reports for the 2011-2020 periods. The analysis technique used panel regression. The result obtained is that there is a significant difference in the level of bank soundness between conventional banks and Islamic banks. Based on a comparison of financial ratio analysis, the financia...

Determining and observing the minimum allowable distance of the marine earth station in motion (M-ESIM) from the shore prevents its destructive interference on the co-frequency shore fixed station. ESIMs are providing broadband Fixed Satellite Services (FSS). This paper studies the parameters involved in determining the minimum allowable distance of the ship from the shore by the interference simulation. The results show that with decreasing the carrier frequency, decreasing latitude or increasing the number of annual passing vessels, this minimum distance increases. In this paper a methodology is presented and simulated to keep constant the minimum allowed distance by adjusting the values of the frequency dependent rejection (FDR). FDR is caused by shifting the M-ESIM frequency band. The minimum distance of 100, 105 and 110Km is evaluated in this paper. In this way, the M-ESIM can be close to the shore as near as the desired distance using the FDR adjustment.

FRANKLIN DELANO ROOSEVELT - FDR (1882-1945) was a Democrat American president for 12 long years, starting at 1933 and up to his expiry in 1945. He gained 86% of the Jewish-American vote. WWII and the HOLOCAUST (1939-1945) occurred fully on his shift. 6 Million Jews were exterminated by the Germans in Europe on FDR's shift in America. FDR was one of the firsts in the Free World to know about the German Gas Chambers in Poland.
So far for the bold irrefutable facts.
What on earth were FDR and his advisers thinking?
Were FDR and the JEWS of AMERICA GULLIBLE?
Was FDR anti-SEMITE?
Did FDR ABANDON the JEWISH VICTIMS?
What the f*** was the #1 persona of the free world thinking???
This 2013 pro-FDR article by one LAURENCE ZUCKERMAN, in THE NATION Magazine goes to advocate FDR's Holocaust mishaps (soft word).
Alas, Zuckerman's Ney to the above questions points to the Yey.
Proper disclosure: My paternal grandparents, aunt, uncle and 400 of my 2nd and 3rd cousins perished in the Holocaust, most of them were from the Warsaw, Poland area, and were gassed in the Treblinka Gas Chambers in the years 1942-1943.
And don't, please don't tell me that FDR did not know in real-time about the mass extermination of Jews in Europe...
