In all, 19 research subjects, with current histories of frequent cocaine use, were exposed to cocaine-related cues to elicit drug craving. We measured the change of occupancy of dopamine at D2-like receptors with positron emission tomography (PET) and inferred a change of intrasynaptic dopamine (endogenous dopamine release), based on the displacement of radiotracer [(11)C]raclopride. Receptor occupancy by dopamine increased significantly in putamen of participants who reported cue-elicited craving compared to those who did not. Further, the intensity of craving was positively correlated with the increase in dopamine receptor occupancy in the putamen. These results provide direct evidence that occupancy of dopamine receptors in human dorsal striatum increased in proportion to subjective craving, presumably because of increased release of intrasynaptic dopamine.

The variation in transposition history of different Ty1-copia group LTR retrotransposons in the species lineages of the Pisum genus has been investigated. A heterogeneous population of Ty1-copia elements was isolated by degenerate PCR and two of these (Tps12 and Tps19) were selected on the basis of their copy number and sequence conservation between closely related species for further in-depth study of their transpositional history in Pisum species. The insertional polymorphism of these elements and the previously characterised PDR1 element was studied by sequence-specific amplification polymorphism (SSAP). Each of these elements reveals a unique transpositional history within 55 diverse Pisum accessions. Phylogenetic trees based on the SSAP data show that SSAP markers for individual elements are able to resolve different species lineages within the Pisum genus. Finally, the SSAP data from all of these retrotransposon markers were combined to reveal a detailed picture of the intra and interspecies relationships within Pisum.

ABSTRACT Though various types of force transducers have been developed over many years, research is continuing through the globe regarding the development of different types of force transducers. The present paper discusses the development of square ring shaped force transducers and their metrological characterization based on the calibration procedures according to standards ISO 376:2004 and IS: 4169-1988 (reaffirmed 2003). The force transducers have been developed as a modification of ring shaped force transducers and strain gauges have been mounted on suitable locations. The force transducers have been studied analytically and computationally. The force transducers are found to have good metrological characteristics and further efforts are being focused on improving the metrological characteristics of force transducers.

ABSTRACT Two low band gap conjugated polymers P1 (alternating phenylenevinylene contg. thiophene and pyrrole rings) and P2 (alternating phenylenevinylene with dithenyl (thienothiadiazole) segments) having optical band gap 1.65 eV and 1.74 eV, resp., were used as electron donor along with the PC70BM as electron acceptor for the fabrication of bulk heterojunction solar cells. The power conversion efficiency (PCE) of BHJ devices based on P1:PC70BM and P2:PC70BM cast from THF solvent is ∼2.84% and 2.34%, resp., which is higher than the BHJ based on PCBM as electron acceptor. The authors have studied the effect of mixed (1-chloronaphthalene (CN)/THF) solvent, modification of PEDOT:PSS layer and inserting of TiO2 layer, on the photovoltaic performance of polymer solar cell. The authors have achieved power conversion efficiency of 5.07% for the polymer solar cells having structure ITO/PEDOT:PSS (modified)/P1:PC70BM (CN/THF cast)/TiO2/Al. A novel small molecule (SM) with a low-band-gap based on acenaphthoquinoxaline was synthesized and characterized. It was soluble in polar solvents such as N,N-dimethylformamide and dimethylacetamide. SM showed broad absorption curves in both solution and thin films with a long-wavelength maximum at 642 nm. The thin film absorption onset was located at 783 nm, which corresponds to an optical band gap of 1.59 eV. SM was blended with PCBM to study the donor-acceptor interactions in the blended film morphology and the photovoltaic response of the bulk heterojunction (BHJ) devices. The cyclic voltammetry measurements of the materials revealed that the HOMO and LUMO levels of SM are well aligned with those of PCBM, allowing efficient photoinduced charge transfer and suitable open circuit voltage, leading to overall power conversion efficiencies (PCEs) of approximately 2.21 and 3.23% for devices with the as-cast and thermally annealed blended layer, respectively. The increase in the PCE with the thermally annealed blend is mainly attributed to the improvement in incident photon to current efficiency (IPCE) and short circuit photocurrent (J(sc)). Thermal annealing leads to an increase in both the crystallinity of the blend and hole mobility, which improves the PCE.

ABSTRACT This paper describes an experiment that allows the quantitative measurement of the CSA−CSA cross-correlation between backbone 15N and 13CO nuclei in uniformly enriched proteins. The CSA−CSA cross-correlation is obtained from the cross-peak intensity ratios of the double- and the zero-quantum components observed with a modified triple-quantum 2D CT-HNCO experiment. In addition, the 1HN−15N/1HN−13CO dipole−dipole cross-correlation was measured without relying on resolved scalar couplings using a complementary quantum 2D CT-HNCO experiment. The cross-correlation rates measured for the protein binase (12.3 kDa) were obtained with high precision but show a surprisingly large range of values. Calculations show that this range is at least partially caused by dynamical processes. The potential use of this information to characterize internal anisotropic motion is discussed.

In this work, addition of OH(-) to one-electron oxidized thymidine (dThd) and thymine nucleotides in basic aqueous glasses is investigated. At pHs ca. 9-10 where the thymine base is largely deprotonated at N3, one-electron oxidation of the thymine base by Cl(2)(•-) at ca. 155 K results in formation of a neutral thyminyl radical, T(-H)·. Assignment to T(-H)· is confirmed by employing (15)N substituted 5'-TMP. At pH ≥ ca. 11.5, formation of the 5-hydroxythymin-6-yl radical, T(5OH)·, is identified as a metastable intermediate produced by OH(-) addition to T(-H)· at C5 at ca. 155 K. Upon further annealing to ca. 170 K, T(5OH)· readily converts to the 6-hydroxythymin-5-yl radical, T(6OH)·. One-electron oxidation of N3-methyl-thymidine (N3-Me-dThd) by Cl(2)(•-) at ca. 155 K produces the cation radical (N3-Me-dThd(•+)) for which we find a pH dependent competition between deprotonation from the methyl group at C5 and addition of OH(-) to C5. At pH 7, the 5-methyl deprotonated species is found; however, at pH ca. 9, N3-Me-dThd(•+) produces T(5OH)· that on annealing up to 180 K forms T(6OH)·. Through use of deuterium substitution at…
