There are significant levels of concern about both the safety assessment and financial evaluation of the whole hydropower system, especially at early project stages. In addition, there is a variety of reliable and accurate methods for analysis, design, and optimization of steel pressure liners in hydropower plants. Several countries have developed specific regulations and codes for the design, installation, and safety evaluation of under-pressure piping, as well as estimates of the potential risks associated with failure. This paper reviews the current methodologies and codes available for design and safety assessment of either unstiffened or stiffened pressure steel liners in hydropower plants. After examining the main guidelines and practical conclusions drawn from recent research in the field of liners subjected to either internal or external pressure, this article then discusses the relevant regulations and codes. The scope of this work is to summarize the advantages, disadvanta...

The Accreditation Board for Engineering and Technology (ABET) has highlighted two key outcomes for students of all accredited engineering programs: the ability to communicate effectively with a range of audiences and the capacity to acquire and apply new knowledge as needed, using appropriate learning strategies. Likewise, in recent years, written exams, assignments, and oral presentations show transmission-skill deficiencies among engineering students. Flipped teaching serves to boost students to meet these outcomes and other competencies: comprehension reading, communication skills, character building, collaborative work, critical thinking, or creativity. So, flipped learning is more than watching videos. This research proposes two evidence-based transferable learning strategies built on a flipped-teaching model and was applied by the authors in engineering courses during the second year of the global pandemic caused by COVID-19: problem-based learning and teamwork assignments. As a consequence of the global health emergency in early 2020, universities had to tackle a sudden shift in their teaching–learning strategies so that the preset competences could be fulfilled. This study presents the learning outcomes of the implemented tasks, student experiences, and feedback, as well as some reflections from the instructors with a holistic perspective of the courses due to the adopted measures and adaptations. Six courses taught at civil engineering degrees of three universities, two from Spain and one from Peru, were analyzed. The teaching and evaluation strategies are described, and some reflections are made by comparing the student’s performance with the previous course. Though the shift to online learning had to be made from day to day, with no time for preparation, the experience has proved that online learning can be beneficial in some aspects and has probably come to stay, although some other aspects are difficult to replace with respect to face-to-face le...

Indigenous beers (chicha) are part of the indigenous culture in Ecuador. The fermentation process of these beers relay probably on microorganisms from: fermenting substrates, environment and human microbiota. We analyzed the microbiota of artisanal beers (including a type of beer produced after chewing boiled cassava) using bacterial culture and 16S-based tag-encoded FLX amplicon pyrosequencing (bTEFAP). Surprisingly, we found that Streptococcus salivarius and Streptococcus mutans (part of the human oral microbiota) where among the most abundant bacteria in chewed cassava and in non-chewed cassava beers. We also demonstrated that S. salivarius and S. mutans (isolated from these beers) could proliferate in cassava mush. Lactobacillus sp. was predominantly present in most types of Ecuadorian chicha.

The aim of this study was to compare the frequency of ERG rearrangement, PTEN deletion, SPINK1 overexpression, and SPOP mutation in prostate cancer in African American and Caucasian men. Dominant tumor nodules from radical prostatectomy specimens of 105 African American men (AAM) were compared with 113 dominant nodules from Caucasian men (CaM). Clinical and pathologic characteristics of the two groups were similar. SPINK1 overexpression was evaluated by immunohistochemistry, ERG rearrangement and PTEN deletion by FISH, and SPOP mutation by Sanger sequencing. ERG rearrangement was identified in 48 of 113 tumors (42.5%) in CaM and 29 of 105 tumors (27.6%) in AAM (P = 0.024). PTEN deletion was seen in 19 of 96 tumors (19.8%) in CaM and 7 of 101 tumors (6.9%) in AAM (P = 0.011). SPINK1 overexpression was present in 9 of 110 tumors (8.2%) in CaM and 25 of 105 tumors (23.4%) in AAM (P = 0.002). SPOP mutation was identified in 8 of 78 (10.3%) tumors in CaM and 4 of 88 (4.5%) tumors in AAM ...

The TMPRSS2–ETS fusion prostate cancers comprise 50–70% of the prostate‐specific antigen (PSA)‐screened hospital‐based prostate cancers examined to date, making it perhaps the most common genetic rearrangement in human cancer. The most common variant involves androgen‐regulated TMPRSS2 and ERG, both located on chromosome 21. Emerging data from our group and others suggests that TMPRSS2–ERG fusion prostate cancer is associated with higher tumour stage and prostate cancer‐specific death. The goal of this study was to determine if this common somatic alteration is associated with a morphological phenotype. We assessed 253 prostate cancer cases for TMPRSS2–ERG fusion status using an ERG break‐apart FISH assay. Blinded to gene fusion status, two reviewers assessed each tumour for presence or absence of eight morphological features. Statistical analysis was performed to look for significant associations between morphological features and TMPRSS2–ERG fusion status. Five morphological featu...

Purpose: More than 1,300,000 prostate needle biopsies are done annually in the United States with up to 16% incidence of isolated high-grade prostatic intraepithelial neoplasia (HGPIN). HGPIN has low predictive value for identifying prostate cancer on subsequent needle biopsies in prostate-specific antigen–screened populations. In contemporary series, prostate cancer is detected in ∼20% of repeat biopsies following a diagnosis of HGPIN. Further, discrete histologic subtypes of HGPIN with clinical implication in management have not been characterized. The TMPRSS2-ERG gene fusion that has recently been described in prostate cancer has also been shown to occur in a subset of HGPIN. This may have significant clinical implications given that TMPRSS2-ERG fusion prostate cancer is associated with a more aggressive clinical course. Experimental Design: In this study, we assessed a series of HGPIN lesions and paired prostate cancer for the presence of TMPRSS2-ERG gene fusion. Results: Fusion...

Purpose: Fusion of the TMPRSS2 prostate-specific gene with the ERG transcription factor is a putatively oncogenic gene rearrangement that is commonly found in prostate cancer tissue from men undergoing prostatectomy. However, the prevalence of the fusion was less common in samples of transurethral resection of the prostate from a Swedish cohort of patients with incidental prostate cancer followed by watchful waiting, raising the question as to whether the high prevalence in prostatectomy specimens reflects selection bias. We sought to determine the prevalence of TMPRSS2-ERG gene fusion among prostate-specific antigen–screened men undergoing prostate biopsy in the United States. Experimental Design: We studied 140 prostate biopsies from the same number of patients for TMPRSS2-ERG fusion status with a fluorescent in situ hybridization assay. One hundred and thirty-four samples (100 cancer and 34 benign) were assessable. Results: ERG gene rearrangement was detected in 46% of prostate b...

Background: This study aimed to evaluate the association of recurrent molecular alterations in prostate cancer, such as ERG rearrangements and phosphatase and tensin homolog gene (PTEN) deletions, with oncologic outcomes in patients with prostate cancer treated with brachytherapy. Methods: Ninety-two men underwent I-125 brachytherapy with a 145 Gy delivered dose between 2000 and 2008. Pretreatment prostate biopsies were analyzed by immunohistochemistry (IHC) and FISH for ERG rearrangement and overexpression, PTEN deletion, and expression loss. Univariable and multivariable Cox-regression analyses evaluated association of ERG and PTEN status with biochemical recurrence (BCR). Results: Within a median follow-up of 73 months, 11% of patients experienced BCR. Of 80 samples with both IHC and FISH performed for ERG, 46 (57.8%) demonstrated rearrangement by FISH and 45 (56.3%) by IHC. Of 77 samples with both IHC and FISH for PTEN, 14 (18.2%) had PTEN deletion by FISH and 22 (28.6%) by IHC....

Enhanced Geothermal Systems (EGS) can boost sustainable development by providing a green energy supply, although they usually require the hydraulic stimulation of the reservoir to increase fluid flow and energy efficiency due to the low rock permeability at the required depths. The injection of fluids for hydraulic stimulation implies several risks, for instance, induced seismicity. In this work, we perform numerical simulations to evaluate the seismic risk in terms of fault reactivation, earthquake magnitude, and rupture propagation. The computational model includes the fully coupled thermo-hydro-mechanical equations and simulates faults as frictional contacts governed by rate-and-state friction laws. We apply our methodology to the Basel EGS project as a continuation of our previous work, employing the same parameters and conditions. Our results demonstrate that permeability stimulation is not only related to induced seismicity but also can induce a thermal decline of the reservoi...

Understanding the risk associated with anthropogenic earthquakes is essential in the development and management of engineering processes and hydraulic infrastructure that may alter pore pressures and stresses at depth. The possibility of earthquakes triggered by reservoir impoundment, ocean tides, and hydrological events at the Earth surface (hydro-seismicity) has been extensively debated. The link between induced seismicity and hydrological events is currently based on statistical correlations rather than on physical mechanisms. Here, we explore the geomechanical conditions that could allow for small pore pressure changes due to reservoir management and sea level changes to propagate to depths that are compatible with earthquake triggering at critically-stressed faults (several kilometers). We consider a damaged fault zone that is embedded in a poroelastic rock matrix, and conduct fully coupled hydromechanical simulations of pressure diffusion and rock deformation. We characterize ...

Geothermal energy has emerged as an alternative to ensure a green energy supply while tackling climate change. Geothermal systems extract the heat stored in the Earth’s crust by warming up water, but the low rock permeability at exploitation depths may require the hydraulic stimulation of the rock fracture network. Enhanced Geothermal Systems (EGS) employ techniques such as hydro-shearing and hydro-fracturing for that purpose, but their use promotes anthropogenic earthquakes induced by the injection or extraction of fluids. This work addresses this problem through developing a computational 3D model to explore fault reactivation and evaluating the potential for earthquake triggering at preexisting geological faults. These are included in the model as frictional contacts that allow the relative displacement between both of its sides, governed by rate-and-state friction laws and fully coupled with thermo-hydro-mechanical equations. We apply our methodology to the Basel project, employ...
